摘  要:目的研究刺芒柄花素对高糖诱导的大鼠H9c2心肌细胞损伤的影响。方法将H9c2心肌细胞分成正常对照组(5.5 mmol·L^(-1)葡萄糖)、高糖模型组(30 mmol·L^(-1)葡萄糖)、刺芒柄花素低剂量(30 mmol·L^(-1)葡萄糖+25 mol·L^(-1)刺芒柄花素)和刺芒柄花素高剂量组(30 mmol·L^(-1)葡萄糖+50 mol·L^(-1)刺芒柄花素)。给药24 h后。通过细胞增殖计数法检测细胞存活率;通过多功能酶标仪测定活性氧(ROS)、超氧化物歧化酶(SOD)活力和乳酸脱氢酶(LDH)水平;用酶联免疫吸附法测定白细胞介素(IL)-1β、IL-6、IL-10和肿瘤坏死因子-α(TNF-α)细胞因子水平;用蛋白质印迹法检测Toll样受体4(TLR4)信号通路蛋白的表达情况。结果正常对照组、高糖模型组、刺芒柄花素低剂量组和刺芒柄花素高剂量组的存活率分别为(100.00±0.10)%,(62.00±0.40)%,(78.00±0.50)%和(94.00±0.20)%,ROS水平分别为1.00±0.04,4.50±0.15,2.17±0.22和1.35±0.43,SOD活力分别为(180.13±0.75),(37.42±0.72),(117.95±0.65)和(152.14±0.25)U·mg^(-1)prot,LDH活力分别为(1409.79±2.38),(2846.44±2.17),(1974.21±1.94)和(1643.72±2.24)U·L^(-1),TLR4蛋白相对表达水平分别为0.03±0.14,0.78±0.11,0.57±0.25和0.24±0.59,差异均有统计学意义(均P<0.05)。与正常对照组相比,高糖模型组中IL-1β、IL-6和TNF-α的水平均显著升高,IL-10水平显著降低(均P<0.05);与高糖模型组相比,刺芒柄花素低、高剂量组中IL-1β、IL-6和TNF-α水平均明显下降,IL-10水平明显提高(均P<0.05),并且存在着剂量依赖关系。结论刺芒柄花素可以改善高糖诱导H9c2心肌细胞损伤,其作用机制可能是通过降低H9c2心肌细胞氧化损伤、抑制细胞凋亡、抑制炎症反应、调控TLR4信号通路来发挥保护作用的。ObjectiveTo investigate the protective effect of formononetin on cell damage and in H9c2 cardiomyocytes induced by high concentration of glucose.Methods H9c2 cardiomyocytes were divided into four groups:control group(5.5 mmol·L^(-1)glucose),high glucose model group(30 mmol·L^(-1)glucose),low(30 mmol·L^(-1)glucose+25 mol·L^(-1)formononetin)and high(30 mmol·L^(-1)glucose+50 mol·L^(-1)formononetin)dose formononetin groups.After treatment with different drug for 24 h,cell counting kit-8 assay was used to determine H9c2 cardiomyocytes cell viability;reactive oxygen species(ROS),superoxide dismutase(SOD)and lactate dehydrogenase(LDH)levels were measured by multi-detection reader;interleukin(IL)-1β,IL-6,IL-10 and tumor necrosis factor-α(TNF-α)levels were measured by enzyme-linked immunosorbent assay.The protein expression of Toll-like receptors-4(TLR4)signaling pathway was determined by Western blot.Results The cells viability of control group,high glucose model group,low and high dose formononetin groups were(100.00±0.10)%,(62.00±0.40)%,(78.00±0.50)%,(94.00±0.20)%;the ROS levels were 1.00±0.04,4.50±0.15,2.17±0.22,1.35±0.43;the SOD viability were(180.13±0.75),(37.42±0.72),(117.95±0.65),(152.14±0.25)U·mg^(-1)prot;the LDH viability were(1409.79±2.38),(2846.44±2.17),(1974.21±1.94),(1643.72±2.24)U·L^(-1);the TLR4 protein expression levels were0.03±0.14,0.78±0.11,0.57±0.25,0.24±0.59,the difference were statistically significant(all P<0.05).Compared with control group,the levels of IL-1β,IL-6 and TNF-αin high glucose model group were significantly increased,and the level of IL-10 was significantly decreased(all P<0.05).Compared with the high glucose model group,the levels of IL-1β,IL-6 and TNF-αin low and high dose formononetin groups were significantly decreased,and the levels of IL-10 were significantly increased(all P<0.05),with dose-dependent.ConclusionFormononetin can alleviate the H9c2 cardiomyocytes damage induced by high concentration of glucose via reducing oxidative damage,inhibiti
