摘 要:为研究鸭坦布苏病毒(DTMUV)NS1蛋白的各项生物信息学特征,本研究利用ExPASY预测分析NS1蛋白的理化性质和亲水性;TMHMM-2.0预测分析NS1蛋白的跨膜区;SignalP 4.1预测分析NS1蛋白的信号肽;SOPMA预测分析NS1蛋白的二级结构;Phyre2构建NS1蛋白的三级结构模型;NetNGlyc 1.0等预测分析NS1蛋白的翻译后修饰位点;IEDB预测分析NS1蛋白的B细胞抗原表位;NetMHCII pan-4.1预测分析NS1蛋白的T细胞抗原表位。结果表明:NS1蛋白为稳定性和亲水性蛋白,由352个氨基酸组成,相对分子质量(Mr)为39628.00,理论等电点为7.97,无跨膜区和信号肽;NS1蛋白具有16个潜在N-糖基化修饰位点、10个潜在O-糖基化修饰位点以及40个潜在磷酸化修饰位点;NS1蛋白具有多个潜在B细胞和T细胞抗原表位。本研究完成了对DTMUV NS1系统性的生物信息学特征分析,为NS1蛋白功能研究以及鸭坦布苏病毒新型疫苗的研发提供依据。To study the bioinformatics characteristics of nonstructural protein 1(NS1)of duck Tembusu virus,in this study that physicochemical properties and hydrophobicity of NS1 protein were analyzed by ExPASY,transmembrane domains of NS1 protein was analyzed by TMHMM-2.0,signal peptides of NS1 protein was analyzed by SignalP 4.1,secondary structures of NS1 protein was analyzed by SOPMA,tertiary structures of NS1 protein was analyzed by Phyre2,post-translational modification sites of NS1 protein was analyzed by NetNGlyc 1.0,the B epitopes of NS1 protein was analyzed by IEDB,the T cell epitopes of NS1 protein was analyzed by NetMHCII pan-4.1.The results showed that NS1 protein was composed of 352 amino acids with relative molecular mass(Mr)of 39628.00 and a theoretical isoelectric point of 7.97.NS1 protein was a stable and hydrophilic protein and has no signal peptide sequences and a potential transmembrane region.NS1 protein contained 16 N-glycosylation sites,10 O-glycosylation sites,and 40 phosphorylation sites.NS1 protein contained multiple potential B cell and T cell epitopes.This study completed systematic bioinformatic analysis of NS1 protein,which provide a basis for the functional study of NS1 protein and the development of duck tambusu virus vaccine.
