摘 要:目的探讨升血小板胶囊对小鼠免疫性血小板减少症(ITP)的治疗作用及免疫调控机制。方法将40只小鼠随机分为对照组、模型组、强的松组、胶囊组各10只。除对照组外,其余三组均用抗血小板抗体于小鼠腹腔内注射,诱导持续性血小板减少成功建立ITP模型。对照组、模型组给予生理盐水灌胃,强的松组、胶囊组分别给予相同体积的强的松溶液、升血小板胶囊溶液灌胃,每日1次,连续8 d。模型维持4周处死动物,计数小鼠外周血小板、骨髓涂片产板巨核细胞,检测小鼠脾脏Tregs细胞构成比、Tregs/CD4值以及小鼠脾脏Tregs细胞Foxp3mRNA表达、Teff细胞IL-2 mRNA表达。结果对照组、模型组、强的松组、胶囊组小鼠外周血小板计数分别为(600.35±145.39)、(123.33±16.24)、(327.66±73.20)、(577.35±76.94)×10^(9)/L,骨髓产板巨核细胞计数分别为(55.05±5.83)、(26.90±2.49)、(66.27±3.58)、(63.45±3.44)个;与模型组比较,胶囊组、强的松组ITP小鼠外周血小板水平升高、产板巨核细胞数增加(P均<0.05)。与模型组比较,胶囊组、强的松组ITP小鼠脾脏Tregs细胞构成比、Tregs/CD4值、Tregs细胞Foxp3mRNA表达量升高,Teff细胞IL-2 mRNA表达量下调,差异均有统计学意义(P均<0.05)。结论升血小板胶囊治疗可以提升ITP小鼠外周血小板计数,其机制可能与其提高脾脏Tregs细胞构成比例、改善机体免疫监视状态有关。Objective To study the therapeutic effect of Sheng Xuexiaoban capsules(SXXCs)on immune thrombocytopenia(ITP)mice and its immune regulation mechanism.Methods Forty mice were randomly divided into the normal group,control group,prednisone group,and SXXCs group,with 10 mice in each group.Except for the normal group,the mice in the other three groups were intraperitoneally injected with anti-platelet antibody to induce persistent thrombocytopenia to establish ITP models successfully.Mice in the normal group and model group were given normal saline through intragastric administration,while mice in the prednisone group and SXXCs group were given the same volume of prednisone solution and SXXCs solution through intragastric administration,once a day,for consecutive 8 days.The models were maintained for 4 weeks and the animals were sacrificed.We assessed changes in platelet counts,megakaryocyte counts,the ratio of Tregs cells in the spleen,the ratio of Treg/CD4,and the expression levels of Foxp3-mRNA in Tregs and IL-2 mRNA of Teff in the spleen.Results The peripheral blood platelet count in the normal group,control group,prednisone group,and SXXCs group was(600.35±145.39),(123.33±16.24),(327.66±73.20),(577.35±76.94)×10^(9)/L,respectively.The number of megakaryocytes in the bone marrow was 55.05±5.83,26.90±2.49,66.27±3.58,and 63.45±3.44.Compared with the model group,ITP mice in the SXXCs group and prednisone group had higher peripheral blood platelets level and platelet-producing megakaryocyte number(all P<0.05),and Treg,Treg/CD4 ratio and Foxp3-mRNA expression level of Tregs cells in spleen increased.The expression of IL-2 mRNA in Teff cells was down-regulated,with statistically significant difference(all P<0.05).Conclusion SXXCs treatment can significantly increase the peripheral blood platelet count in the ITP model mice,and its mechanism may be related to the increase of the proportion of Tregs cells in spleen and the improvement of immune surveillance.
