摘  要:目的明确常见益生菌(屎肠球菌、枯草芽孢杆菌、粪肠球菌、蜡样芽孢杆菌)对V583杀伤作用的最适条件及分子机制。方法利用细菌共培养技术,筛选对V583具有明显杀伤作用的益生菌;按照倍比稀释的方法,比较6~24 h内V583杀伤效率;通过基因组学比较的方式,明确不同杀伤效率的细菌基因组学差异,发现与V583杀伤密切相关的潜在分子机制。结果粪肠球菌对V583具有最佳杀伤作用;粪肠球菌与V583等量时可在6 h内,有效杀伤70%以上;比较基因组学研究表明:粪肠球菌的溶血素基因以及毒素-抗毒素系统(TA系统)与杀伤密切相关,可能发挥着关键作用。结论本研究通过比较不同益生菌对粪肠球菌V583的杀伤开展研究,发现了对V583发挥杀伤作用的最小菌量及可能的分子机制,对V583临床防治过程中益生菌的开发,有重要的指导作用。OBJECTIVE To determine the optimal conditions and molecular mechanism of killing V583 by common probiotics(Enterococcus faecium,Bacillus subtilis,Enterococcus faecalis,Bacillus cereus).METHODS Probiotics that had obvious killing effect on V583 were screened by bacterial co-cultivation technology.The killing efficiency of V583 between within 6 and 24 hours were compared according to the method of multiple dilution.The differences in bacterial genomics between bacteria of different killing efficiencies were compared and the mechanism of V583 killing was explored.RESULTS E.faecalis has the best killing effect on V583;E.faecalis can effectively kill more than 70%of the V583 strains within 6 hours when they were equally prepared.Comparative genomics analysis showed that hemolysin gene and toxin-antitoxin system(TA system)of E.faecalis were closely related to the killing role of the strain.CONCLUSION In this study,the killing effect of various E.faecalis isolated from human body is analyzed and the molecular mechanism of the effect on V583 in the minimum bacteria is found,which plays a certain guiding role in the development of probiotics for the prevention and treatment of V583 in clinic.
