半夏泻心汤人含药血清对Hp感染GES-1细胞凋亡及XIAP表达的影响

摘　　要：目的探究半夏泻心汤人含药血清对幽门螺杆菌(Hp)感染人胃黏膜上皮细胞GES-1凋亡及X连锁凋亡抑制蛋白(XIAP)表达的影响及其作用机制。方法临床收集医院2018年6月-2020年8月健康志愿者血清和Hp相关消化性溃疡患者分别给予半夏泻心汤、标准三联疗法(雷贝拉唑钠肠溶片+克拉霉素+甲硝唑)治疗前、后血清,将GES-1细胞分为空白组、Hp感染组、用药前组、半夏泻心汤组、标准三联组,除空白组外均体外接种Hp建立Hp感染模型,血清干预24 h后,MTT法检测细胞活力,流式细胞术检测细胞凋亡;检测细胞超氧化物歧化酶(SOD)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白细胞介素-8(IL-8)水平;检测X连锁凋亡抑制蛋白(XIAP)、天冬氨酸蛋白水解酶3(Caspase-3)、核因子κB抑制蛋白α(IKBα)、核因子κB(NF-κB)p65蛋白表达、mRNA相对表达量。结果与Hp感染组比较,半夏泻心汤组、标准三联组细胞增殖抑制率、细胞凋亡率、细胞内SOD水平降低,细胞内MDA、TNF-α、IL-1β、IL-8水平升高,BAX蛋白表达水平上调,BCL-2蛋白表达水平下调,XIAP蛋白表达水平和mRNA相对表达量上调,IKBα、NF-κB p65蛋白表达水平和mRNA相对表达量下调,cleaved caspase-3/caspase-3比率降低,caspase-3 mRNA相对表达量下调,均有统计学差异(P<0.05)。结论半夏泻心汤能抑制Hp感染诱导的GES-1细胞凋亡,降低其炎性反应和氧化应激,其作用机制可能与上调XIAP表达,抑制NF-κB信号通路有关。OBJECTIVE To explore the effect and mechanism of serum of patients treated with Banxia Xiexin decoction on Helicobacter pylori(Hp)infected human gastric epithelial mucosal cells(GES-1)apoptosis and expression of X-linked inhibitor of apoptosis protein(XIAP).METHODS Serum of healthy volunteers and patients with Hp-associated peptic ulcer before and after treatment with Banxia Xiexin decoction or standard triple therapy(rabeprazole sodium enteric-coated tablets+clarithromycin+metronidazole)in the hospital between Jun 2018 and Aug 2020 were collected.The GES-1 cells were divided into the blank group,Hp infection group,pre-medication group,Banxia Xiexin decoction group and standard triple group.The above groups except the blank group were inoculated with Hp in vitro to establish Hp infection model.After 24 h of serum intervention,cell viability was detected by MTT and cell apoptosis was detected by flow cytometry.The levels of superoxide dismutase(SOD),malondialdehyde(MDA),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-8(IL-8)were detected.The protein and mRNA expression of X-linked inhibitor of apoptosis protein(XIAP),aspartic acid proteolytic enzyme 3(Caspase-3),nuclear factor-κB inhibitory proteinα(IKBα)and nuclear factor-κB(NF-κB)p65 were detected.RESULTS Compared with the Hp infection group,the cell proliferation inhibition rate in the Banxia Xiexin decoction group and the standard triple group was significantly reduced(P<0.05);the apoptosis rate was significantly decreased(P<0.05);the intracellular SOD level was significantly lowered(P<0.05),while the levels of intracellular MDA,TNF-α,IL-1βand IL-8 were significantly increased(P<0.05);the BAX protein expression level was significantly up-regulated(P<0.05)while the BCL-2 protein expression level was significantly down-regulated(P<0.05);the protein expression level and mRNA relative expression of XIAP were significantly up-regulated(P<0.05)while the protein expression levels and mRNA relative expression levels of IKBαand NF-κB p6
