摘  要:为克服紫荆属(Cercis Linn.)种间杂交种子后期发育障碍,提高杂交后代成活率,缩短育种周期,本实验以紫荆属的湖北紫荆和紫荆杂交的未成熟胚为外植体材料,研究不同消毒方式、不同胚龄对杂交胚萌发的影响以及筛选最适诱导、增殖及生根培养基。结果表明:消毒方式用体积分数为0.1%的HgCl2溶液处理5 min的效果最佳;存活率为76.67%。在授粉后120 d时采样,胚的萌发率(66.3%)最高。胚拯救最适宜的诱导培养基:MS+3.0 mg·L^(-1)6-BA+0.05 mg·L^(-1)NAA+0.05 mg·L^(-1)GA+150 mg·L^(-1)AC,诱导率为95%;最佳增殖培养基:MS+1.5 mg·L^(-1)KT+1.5 mg·L^(-1)6-BA+0.05 mg·L^(-1)NAA+150 mg·L^(-1)AC,增殖系数为4.79;最佳生根培养基:1/2MS+0.5 mg·L^(-1)NAA+0.1 mg·L^(-1)VH+150 mg·L^(-1)AC,生根率为81.67%。下地炼苗后的移栽成活率为87.53%。本实验旨在建立有效的胚拯救体系,为提高紫荆属种间远缘杂交成功率提供有效技术,同时为紫荆属种间杂交育种提供新的种质资源。In order to overcome the late developmental obstacles of interspecific hybrid seeds of Cercis Linn.,increase the survival rate of interspecific hybrids of Cercis Linn.and shorten the breeding cycle,immature embryos of C.glabra×C.chinensis were used to investigate the effect of different disinfection methods and different embryo ago on the germination of hybrid embryos and to choose out the optimun induction,proliferation and rooting media.The results showed that the best disinfection method was to disinfect the seed for 5 min with 0.1%HgCl2,which the survival rates were 76.67%.The optimal sampling times were 120 days after pollination,with the highest developed embryo rate(66.3%).MS+3.0 mg·L^(-1)6-BA+0.05 mg·L^(-1)NAA+0.05 mg·L^(-1)GA3+150 mg·L^(-1)AC was the best medium for embryo rescue,the induction rate of embryo was 95%.The optimal medium for proliferation was MS+1.5 mg·L^(-1)KT+1.5 mg·L^(-1)6-BA+0.05 mg·L^(-1)NAA+150 mg·L^(-1)AC,the proliferation coefficient was 4.79.And 1/2MS+0.5 mg·L^(-1)NAA+0.1mg·L^(-1)VH+150 mg·L^(-1)AC for rooting was best with the rate of 81.67%.The survival rate of transplanting after hardening in the ground was 87.53%.The purpose of this experiment is to establish an effective embryo rescue system,to provide an effective technology for improving the success rate of distant hybridization among Cercis Linn.,and to provide new germplasm resources for Cercis Linn.interspecific hybrid breeding.
