摘 要:为了探究赪桐潜在防治奶牛乳房炎的作用机制,试验基于网络药理学,利用TCMSP数据库收集赪桐潜在抗奶牛乳房炎致病菌活性成分,在GeneCards数据库中收集与奶牛乳房炎致病菌相关的作用靶点。利用Cytoscape 3.8.2软件绘制赪桐活性成分与致病菌靶点网络图,通过将筛选到的致病菌靶点导入STRING数据库构建致病菌靶点相互作用网络,最后利用DAVID、Metascape数据库对各靶点进行GO功能注释和KEGG富集分析。结果表明:共得到赪桐潜在抗奶牛乳房炎活性成分11个,对应致病菌靶点86个;共得到97个节点,174条相互作用关系;去除游离的病菌靶点后,最终得到31个关键作用靶点。GO功能注释得到312条基因数量较大的功能注释,主要涉及细胞膜、原子核、蛋白质结合、基于转录正向调控过程、信号转导过程等。KEGG富集分析主要涉及蛋白结合通路、酶结合通路、ATP结合通路等。说明基于网络药理学预测,赪桐可通过多种活性成分作用于不同靶点,并通过调控炎症反应、氧化还原反应等过程达到防治奶牛乳房炎的目的。In order to investigate the potential mechanism of Clerodendrum japonicum against cow mastitis, based on network pharmacology, experiment used TCMSP database to collect the potential active ingredients of Clerodendrum japonicum against mastitis-causing bacteria in dairy cows, and the GeneCards database to collect the action targets associated with mastitis-causing bacteria in dairy cows. Cytoscape 3.8.2 software was used to map the network of active ingredients and pathogenic targets of Clerodendrum japonicum, and the pathogenic target interaction network was constructed by importing the screened pathogenic targets into STRING database, and finally GO function annotation and KEGG enrichment analysis were performed for each target by using DAVID and Metascape databases. The results showed that a total of 11 potential anti-mastitis active ingredients of Clerodendrum japonicum were obtained, corresponding to 86 targets of pathogenic bacteria. GO functional annotation yielded a large number of functional annotations for 312 genes, mainly involving cell membrane, nucleus, protein binding, positive transcription-based regulatory processes, and signaling processes. The KEGG enrichment analysis mainly involved protein binding pathway, enzyme binding pathway, and ATP binding pathway. The results indicated that based on the network pharmacology it was predicted that Clerodendrum japonicum could act on different targets through multiple active ingredients to achieve the prevention and treatment of cow mastitis by regulating the inflammatory response, redox reaction and other processes.
