HPLC法测定蒙药蓝刺头中7种有机酸的含量

摘　　要：本研究建立同时测定蒙药蓝刺头中7种有机酸类成分含量的方法。采用Waters HSS T3 C18色谱柱(4.6 mm 250 mm,5 m),流动相为乙腈(A)-0.08%三氟乙酸水溶液(B),梯度洗脱,流速1.0 mL/min,检测波长为325 nm,柱温为室温。同时测定蓝刺头中5-咖啡酰奎尼酸、绿原酸、咖啡酸、芦丁、3,4-二咖啡酰奎尼酸、3,5-O-二咖啡酰基奎宁酸和4,5-O-二咖啡酰奎宁酸等7个有机酸的含量。5-咖啡酰奎尼酸在0.936~93.6μg/mL、绿原酸在1.021~102.1μg/mL、咖啡酸在0.952~95.2μg/mL、芦丁在0.932~93.2μg/mL、3,4-二咖啡酰奎尼酸在0.928~92.8μg/mL、3,5-O-二咖啡酰基奎宁酸在0.964~96.4μg/mL、4,5-O-二咖啡酰奎宁酸在0.927~92.7μg/mL范围内具有良好的线性关系。7种成分的回收率分别在97.89%~99.67%,RSD分别在1.54%~2.11%,表明回收率良好。采用本方法可同时测定蓝刺头中7个有机酸类成分的含量,具有可行性与专属性。The study established a quality to determine the contents of seven organic acids in mongolian medicine Echinops latifolius simultaneously.An HPLC method was established with a Waters HSS T3 C18 column(4.6 mm 250 mm,5 m)with a gradient elution using0.08%trifluoroacetic acid solution-acetonitrile as the mobile phase.The flow rate was 1 mL/min,the detection wavelength was 325 nm and the column temperature was room temperature.The contents of neochlorogenic acid,chlorogenic acid,caffeic acid,rutin,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C in mongolian medicine Echinops latifolius were determined at the same time.Neochlorogenic acid in the range of 0.936~93.6μg/mL,chlorogenic acid is 1.021~102.1μg/mL,caffeic acid at 0.952~95.2μg/mL,rutin at0.932~93.2μg/mL,isochlorogenic acid B in the range of 0.928~92.8μg/mL,isochlorogenic acid A in the range of 0.964~96.4μg/mL,isochlorogenic acid C in the range of 0.927~92.7μg/mL.There is a good linear relationship,and recoveries of the seven components were97.89%~99.67%respectively,and the RSD was 1.54%~2.11%respectively,indicating that the rate of recovery were good.Simultaneously determine the contents of seven organic acids in mongolian medicine Echinops latifolius Tausch showed a favorable feasibility and specificity,which can be used to determine the content of seven organic acids in E.latifolius.
