摘 要:为研究影响棉籽相关产品中游离棉酚检测的因素,本实验通过苯胺法和高效液相色谱法检测脱酚棉籽蛋白游离棉酚含量的差异;利用苯胺法检测添加棉酚类似物、油脂氧化、黄酮类物质和糖类对游离棉酚的影响。结果表明:高效液相法检测脱酚棉籽蛋白的游离棉酚比苯胺法的结果显著偏低(乙醇萃取偏低85%,丙酮萃取偏低89%,P <0.05);棉酚氧化后,高效液相法比苯胺法结果偏低30.15%(P <0.05);油脂氧化会使游离棉酚检测结果增加6.35倍。由此可见,苯胺法比高效液相色谱法检测游离棉酚结果偏高,棉酚类似物、油脂氧化、黄酮类和还原糖类物质都能影响苯胺法检测游离棉酚。To study the factors affecting the detection of free gossypol in cottonseed-related products,the difference in free gossypol content of degossypolled cottonseed protein was detected by aniline method and high performance liquid chromatography;The effect of adding gossypol analogs,oil oxidation,flavonoids and sugars on free gossypol was detected by aniline method.The results showed that the free gossypol in the detection of degossypolled cottonseed protein by the HPLC method was much lower than that of the aniline method(the ethanol extraction was 85% lower,and the acetone extraction was 89% lower,P < 0.05);After the oxidation of gossypol,the HPLC method was better than aniline The method result was 30.15% lower(P <0.05);The oxidation of oil increased the test result of free gossypol by 6.35 times.It could be seen that the aniline method was higher than the high performance liquid chromatography method in the detection of free gossypol.Gossypol analogs,oil oxidation,flavonoids and reducing sugars could all affect the detection of free gossypol by the aniline method.
