摘 要:目的 探究益心泰总黄酮对H9c2心肌细胞代谢的影响及其潜在机制。方法 用去甲肾上腺素(NE)诱导H9c2细胞损伤模型。实验设立空白组、模型组、对照组和低、中、高剂量实验组。低、中、高剂量实验组分别给予350μmol·L^(-1)NE和20,50和100μg·mL^(-1)益心泰总黄酮处置;对照组给予350μmol·L^(-1)NE和400μmol·L^(-1)盐酸曲美他嗪处置;模型组给予350μmol·L^(-1)NE处置;空白组给予DMEM处置。用CCK-8法检测H9c2心肌细胞存活率;用酶联免疫吸附试验法检测细胞外液中腺苷三磷酸(ATP)、单磷酸腺苷(AMP)及游离脂肪酸(FFA)的含量;用Western blot法检测过氧化物酶体增殖物激活受体α(PPARα)和过氧化物酶体增殖物激活受体γ辅活化因子1α(PGC-1α)的表达水平。结果 中、高剂量实验组和对照组、模型组的心肌细胞存活率分别为(81.50±2.72)%,(75.71±1.39)%,(91.45±1.51)%和(68.27±2.20)%;ATP含量分别为(1.18±0.04),(1.32±0.10),(1.41±0.15)和(1.06±0.06)μmol;FFA含量分别为(0.53±0.06),(0.48±0.04),(0.45±0.03)和(0.67±0.07)μmol,高剂量实验组的上述指标与模型组比较,差异均有统计学意义(P<0.01,P<0.05)。低、高剂量实验组和对照组、模型组的PPARα蛋白相对表达水平分别为1.11±0.02,1.01±0.02,0.98±0.01和1.20±0.02;PGC-1α蛋白相对表达水平分别为0.38±0.01,0.49±0.01,0.45±0.02和0.37±0.01,低、高剂量实验组的上述指标与模型组比较,差异均有统计学意义(均P<0.01)。结论 益心泰总黄酮对NE诱导的H9c2心肌细胞损伤有保护作用,改善细胞心肌损伤的能量代谢,其作用机制可能与抑制PPARα蛋白的表达、增强PGC-1α蛋白的表达有关。Objective To explore the effect of Yixintai total flavonoids on H9c2 myocardial energy metabolism and its potential mechanism.Methods Norepinephrine (NE) was used to induce H9c2 cell injury model,and blank group,model group,control group and experimental-L,-M,-H groups were established.Experimental-L,-M,-H groups were given 350μmol·L^(-1)NE and 20,50 and 100μg·m L^(-1)of Yixintai total flavonoids respectively;control group was given 350μmol·L^(-1)NE and 400μmol·L^(-1)trimetazidine hydrochloride;model group was treated with 350μmol·L^(-1)NE;blank group was treated with DMEM.The survival rate of H9c2 cardiomyocytes was detected by CCK-8 method;the contents of adenosine triphosphate (ATP),adenosine monophosphate (AMP) and free fatty acid (FFA) in extracellular fluid were detected by enzyme-linked immunosorbent assay;Western blot was used to detect expression levels of peroxisome proliferators-activated receptorα(PPARα) and peroxisome proliferator-activated receptor-γ coactivator 1α (PGC-1α).Results The survival rates of cardiomyocytes in experimental-M,-H groups,control group and model group were (81.50±2.72)%,(75.71±1.39)%,(91.45±1.51)%and (68.27±2.20)%;the ATP contents were (1.18±0.04),(1.32±0.10),(1.41±0.15) and (1.06±0.06)μmol;the FFA contents were (0.53±0.06),(0.48±0.04),(0.45±0.03) and (0.67±0.07)μmol.Compared with model group,the above indexes in experimental-H group were significantly different (P<0.01,P<0.05).The relative expression levels of PPARα protein in the experimental-L,-H groups,control group and model group were 1.11±0.02,1.01±0.02,0.98±0.01 and 1.20±0.02;the relative expression levels of PGC-1α protein were 0.38±0.01,0.49±0.01,0.45±0.02 and 0.37±0.01,and the differences were statistically significant between experimental-L,-H groups and model group (all P<0.01).Conclusion Yixintai total flavonoids have a protective effect on NE-induced H9c2 myocardial cell injury and improve the energy metabolism of myocardial injury.Its mechanism may be related to inhibitin
