摘 要:目的 研究槲皮素对婴幼儿血管瘤内皮细胞凋亡的影响和机制。方法 分离婴幼儿血管瘤内皮细胞,分成对照组、实验低剂量组(80μmol·L^(-1)槲皮素)、实验中剂量组(160μmol·L^(-1)槲皮素)、实验高剂量组(320μmol·L^(-1)槲皮素)、实验高剂量+pcDNA组(转染阴性对照载体,320μmol·L^(-1)槲皮素)、实验高剂量+高迁移率蛋白1(HMGB1)组(转染HMGB1过表达载体,320μmol·L^(-1)槲皮素处理)。用细胞计数试剂盒-8(CCK-8)分析细胞增殖情况,用流式细胞术分析细胞凋亡情况,用蛋白质印迹法检测HMGB1、剪切的胱天蛋白酶3(C-caspase-3)、CCAAT/增强子结合蛋白同源蛋白(CHOP)、糖调节蛋白78(GRP78)蛋白表达。结果 对照组、实验低剂量组、实验中剂量组、实验高剂量组、实验高剂量+pcDNA组、实验高剂量+HMGB1组细胞中HMGB1蛋白表达水平分别为0.89±0.09,0.76±0.08,0.63±0.06,0.55±0.05,0.56±0.08和0.78±0.07;细胞存活率分别为(100.00±12.54)%,(88.51±8.66)%,(74.02±6.94)%,(62.81±8.76)%,(64.12±5.21)%和(83.02±7.15)%;细胞凋亡率分别为(3.85±0.47)%,(6.32±0.84)%,(8.85±1.20)%,(13.95±1.47)%,(14.01±1.63)%和(7.82±0.92)%;C-caspase-3蛋白表达水平分别为0.26±0.02,0.42±0.05,0.49±0.06,0.67±0.08,0.69±0.06和0.44±0.05;CHOP蛋白表达水平分别为0.22±0.03,0.29±0.04,0.42±0.05,0.62±0.06,0.60±0.07和0.40±0.03;GRP78蛋白表达水平分别为0.33±0.04,0.45±0.04,0.53±0.06,0.66±0.07,0.67±0.09和0.42±0.06,上述指标,实验低、中、高剂量组与对照组相比,差异均有统计学意义(均P<0.05);实验高剂量+HMGB1组和实验高剂量+pcDNA组比,差异均有统计学意义(均P<0.05)。结论 槲皮素下调HMGB1激活内质网应激途径诱导婴幼儿血管瘤内皮细胞凋亡。Objective To study the effect and mechanism of quercetin on apoptosis in endothelial cells of infantile hemangioma. Methods The endothelial cells of infantile hemangioma were isolated and divided into control group, experimental low-dose group(80 μmol·L^(-1)quercetin treatment), experimental medium-dose group(160 μmol·L^(-1)quercetin treatment), experimental high-dose group(320 μmol·L^(-1)quercetin treatment), experimental high-dose + pcDNA group(transfected with negative control vector, 320 μmol·L^(-1)quercetin treatment), experimental high-dose + high mobility group box-1(HMGB1) group(transfected with HMGB1 overexpression vector, 320 μmol·L^(-1)quercetin treatment). Cell counting kit-8( CCK-8) experiment was used to analyze cell proliferation,flow cytometry was used to analyze cell apoptosis,Western blot was used to analyze HMGB1,cleaved cysteine-containing aspartic acid Cleaved cysteinyl aspartate specific proteinase 3( C-caspase-3),CCAAT/enhancer-binding protein C/EBP( CHOP),glucose regulated protein 78( GRP78) protein expression.
