贵州都匀仙女虾的分子鉴定

摘  要:2021年9月在贵州省都匀市甲登村一池塘内发现一种仙女虾,采用PCR扩增技术和DNA测序技术对该仙女虾进行鉴定。结果显示,都匀仙女虾COI、16S rRNA基因序列与鹄沼枝额虫(Branchinella kugenumaensis)相似度分别为98.71%、98.96%。结合GenBank中无背甲目部分科物种的同源序列,开展遗传距离和系统发育关系分析,基于COI、16S rRNA基因遗传距离显示,都匀仙女虾与鹄沼枝额虫的遗传距离分别为0.017、0.000。系统发育树显示,都匀仙女虾与鹄沼枝额虫聚在一起形成单支系,COI-based树和16S rRNA-based树的支持率分别为100%和99%。鉴定结果表明,贵州省都匀市甲登村的仙女虾是钗额虫科(Thamnocephalidae)枝额虫属(Branchinella)的鹄沼枝额虫。In September 2021,a fairy shrimp was found in a pond in Jiadeng Village,Duyun City,Guizhou Province,and the fairy shrimp was identified by PCR amplification and DNA sequencing.The results showed that the sequence similarity of the COI and 16S rRNA genes between Duyun fairy shrimp and Branchinella kugenumaensis was 98.71%and 98.96%,respectively.Combined with the homologous sequences of some species in GenBank,the genetic distance and phylogenetic relationship were analyzed.Based on the genetic distance of COI and 16S rRNA genes,the genetic distances between Duyun fairy shrimp and B.kugenumaensis were 0.017 and 0.000,respectively.The phylogenetic tree showed that the Duyun fairy shrimp and B.kugenumaensis clustered together,which formed a single clade.Moreover,the support rates of the COI-based tree and 16S rRNA-based tree were 100%and 99%,respectively.These results indicated that the fairy shrimp in Duyun City was a B.kugenumaensis belonging to the genus Branchinella of the family Thamnocephalidae.
