摘  要:目的:探讨不同低氧训练模式能否通过激活小鼠腹股沟皮下脂肪组织的脂噬作用调控脂代谢。方法:将40只健康C57BL/6小鼠分为对照组(C组),运动组(E组),低氧组(H组),低住高练组(LoHi组),高住低练(HiLo组)五组,每组8只。E组进行跑台运动,运动方案为:25 m/min,1 h/d,5 d/周;H组每天8到18点在13.6%低氧浓度下进行低氧暴露;LoHi组在常氧(21%)氧浓度下生活,在13.6%低氧浓度下进行运动;HiLo组每天8点~16点在氧气浓度为13.6%的低氧帐篷下进行低氧暴露,每次低氧暴露结束后进行常氧环境下的运动,LoHi组与HiLo组的运动方案与E组一致。6周后采用Elisa法测血清甘油三酯水平,苏木精-伊红(HE)染色观察小鼠脂肪细胞,免疫荧光共定位观察自噬关键因子的共定位情况,Western blot等方法检测微管相关蛋白1轻链3的比值(LC3Ⅱ/Ⅰ)、溶酶体相关2型蛋白(LAMP-2)、选择性自噬接头蛋白(p62)、BECN1基因编码蛋白(Beclin1)等自噬相关蛋白表达水平。结果:(1)与C组相比,LoHi组的体重增长显著减少(P<0.05)。(2)与C组相比,E组、H组、LoHi组、HiLo组的小鼠血清中甘油三酯水平显著降低(P<0.01)。(3)HE染色观察到与C组相比,E组、H组、LoHi组、HiLo组的脂肪细胞直径缩小,数量增多。(4)与C组相比,E组、H组、LoHi组、HiLo组的脂肪细胞中的橙黄色斑点增多,且四组的共定位系数也显著提高(P<0.01);此外,E组的共定位系数显著高于H组(P<0.01);LoHi组的共定位系数也显著高于E组和H组(P<0.01)。(5)与C组相比,E组、H组、LoHi组、HiLo组LC3Ⅱ/Ⅰ、LAMP-2蛋白表达显著升高(P<0.01、P<0.05)。此外,LoHi组LC3Ⅱ/Ⅰ的比值显著大于H组(P<0.01),LoHi组LAMP-2的蛋白表达显著高于E组(P<0.05)。与C组相比,LoHi组、HiLo组Beclin1蛋白表达水平显著提高(P<0.01);E组、H组、LoHi组、HiLo组四组之间Beclin1蛋白表达水平未见显著差异(P>0.05)。与C组相比,E组、LoHi组、HiLo组p62的蛋白表达显著降Objective To investigate whether different hypoxic training modes can regulate lipid metabolism by activating lipophagy in the subcutaneous adipose tissue in the groin of mice. Methods Forty healthy C57BL/6 mice were randomly divided into a control group(C,n=8),an exercise group(E,n=8),a hypoxia group(H,n=8),a low-living/high-training group(LoHi,n=8),and high-living/lowtraining group(HiLo,n=8). Group E completed daily 60-minute treadmill training at a speed of 25m/min, five days a week for six weeks, while the group H was exposed to the environment with13.6% oxygen between 8 and 18 o’clock every day. Meanwhile,the LoHi group lived in the environment with normoxic oxygen concentration(21%) and exercised in the environment with 13.6% hypoxic concentration,while the HiLo group was exposed to the oxygen concentration of 13.6% from 8:00 to16:00 every day,and exercised in the normal oxygen environment after the hypoxia exposure. The exercise program of group LoHi and HiLo was the same with that of group E. After six weeks of intervention,the enzyme-linked immuno sorbent assay and hematoxylin-eosin(HE) staining were conducted to observe the serum triglyceride level and adipocytes. Immunofluorescence co-localization was employed to observe the co-localization of key autophagy factors. Moreover,Western blotting was used to detect the expression of microtubule-associated protein 1 light chain 3 ratio(LC3Ⅱ/Ⅰ),lysosomal associated protein type 2(LAMP-2),selective autophagy adaptor protein(p62),BECN1 gene encoding protein(Beclin1) and other autophagy-related proteins. Results(1) The body weight gain decreased significantly in the LoHi group compared to group C(P<0.05).(2) Compared with group C,the average serum triglyceride level decreased significantly in the other 4 groups(P<0.01).(3) HE staining showed that the adipocytes of group E,H,LoHi and HiLo decreased in diameter but increased in number compared with group C.(4) Moreover,the yellow spots in adipocytes of group E,H,LoHi and HiLo were significantly more than gro
