摘 要:“垂衣裳而天下治”,服饰在中华文明发展中起着重要作用,而丝织品作为最珍贵的纺织品之一,更是有着源远流长的历史。然而历经千百年的降解和老化,丝织品文物在出土时已难以辨别形貌,对后续取样鉴别及保护造成了巨大的困难。为解决这一现场检测难题,本文通过设计丝素蛋白单克隆抗体,结合胶体金免疫层析技术制备了桑蚕丝素蛋白快速检测试纸。该试纸可以专一识别桑蚕丝素蛋白,其最低检测限可以达到0.5μg/mL,为考古现场丝绸文物的快速检测提供了一种灵敏、精确、高效的分析手段。Silk is a symbol of ancient Chinese culture.This communication route between the East and the West in Eurasia was named the“Silk Road”.Along the Silk Road,great quantities of splendid Chinese silk spread to the West.There were a great number of silk fabrics that had been unearthed along the Silk Road.They came from different places and had a long history.The excavation of these cultural relics is of great value to the silk origin and development of China’s textile technology and the study of ancient human civilization.However,silk fabrics,which are mainly composed of sericin(a linear and water-soluble protein)and fibroin(a globular and water-soluble protein),are extremely vulnerable to the influences of the burial environment.Many exogenous factors,such as temperature,humidity,microorganisms,environmental acidity and alkalinity,may cause the damage and degradation of silk fabrics,making it difficult to identify the shape of silk relics when unearthed,and also causing difficulties for subsequent sampling and identification.In recent years,immunochromatographic technology has been gradually applied to the archaeological field due to its advantages of quickness,convenience,high specificity and sensitivity.Therefore,the preparation of relevant immunochromatographic assay strips has great significance for the rapid detection of textiles in archaeological sites.In order to solve the problems about the rapid detection at archaeological sites,an assay strip was developed for the rapid detection of Bombyx mori silk fibroin based on the immunological detection combined with membrane technology,using colloidal gold as a tracer.If silk fibroin is present in the sample,it will first bind with colloidal gold-labeled antibody and compete with the antigen formation on the T-line,giving a positive result on the assay strip.Otherwise,the strip will show a negative result.First,a monoclonal antibody against Bombyx mori silk fibroin was prepared,providing the foundation for the specific binding between antigen and antibody.The
