摘 要:目的研究橙皮素(HSP)对高脂血症(HLP)大鼠的血管功能障碍以及血管内皮损伤的保护作用。方法将50只大鼠随机分为对照组、模型组、低剂量实验组、中剂量实验组和高剂量实验组,每组10只。除对照组外其余各组均以高脂饲料喂养法制备高脂大鼠模型。4周后,低、中、高剂量实验组分别灌胃50、100、150 mg·kg^(-1)·d^(-1)HSP,对照组和模型组灌胃等剂量0.5%羧甲基纤维素钠(CMC-Na)。各组均灌胃4周,每日1次。4周后腹主动脉取血。以酶联免疫吸附(ELISA)法测定大鼠血脂总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、血管紧张素Ⅱ(Ang-Ⅱ)、血管内皮生长因子(VEGF)、前列环素2(PGI_(2))和血栓收缩素2(TXA_(2));以血管张力法测定HSP对冠状动脉舒张功能的影响。结果对照组、模型组和低、中、高剂量实验组的TC水平分别为(2.15±0.66),(6.38±1.31),(4.05±0.67),(3.98±1.20)和(3.06±0.92)mmol·L^(-1);这5组的TG水平分别为(0.89±0.17),(1.44±0.21),(1.45±0.23),(1.15±0.13)和(0.92±0.27)mmol·L^(-1);这5组的LDL-C水平分别为(0.62±0.12),(1.47±0.37),(1.33±0.35),(1.23±0.36)和(1.01±0.29)mmol·L^(-1);这5组的HDL-C水平分别为(1.34±0.15),(1.30±0.27),(1.39±0.18),(1.36±0.19)和(1.41±0.11)mmol·L^(-1);这5组的Ang-Ⅱ水平分别为(422.19±12.27),(553.79±13.17),(551.15±11.57),(533.00±11.69)和(496.00±16.67)pg·mL^(-1);这5组的VEGF水平分别为(184.81±2.45),(234.40±3.15),(226.00±4.87),(209.87±4.94)和(196.59±5.41)pg·mL^(-1);这5组的PGI2水平分别为(67.40±5.76),(86.03±5.02),(86.07±4.15),(70.53±7.03)和(68.59±5.86)pg·mL^(-1);这5组的TXA2水平分别为(101.44±5.47),(129.41±6.87),(126.50±5.27),(121.31±5.77)和(114.21±8.09)pg·mL^(-1);这5组对乙酰胆碱(ACh)的最大舒张率分别为(52.57±2.07)%,(32.12±1.93)%,(34.45±2.84)%,(42.97±1.75)%和(50.25±1.28)%。上述指标,模型组与对照组比较,差异均有统计学意义(均Objective To study the protective effect of hesperidin(HSP)on vascular dysfunction and vascular endothelial injury in hyperlipidemia(HLP)rats.Methods Hyperlipidemia model was established by high fat feeding.Fifty male SD rats were randomly divided into control group,model group,experimental-L group,experimental-M group and experimental-H group.The control group was fed with normal diet and other groups were fed with high-fat diet.After 4 weeks,the control group and the model group were gavaged with 0.5%carboxymethylcellulose sodium(CMC-Na),and the other groups were gavaged with HSP.After the experiment,blood samples were taken from the abdominal aorta.Total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C),angiotensinⅡ(Ang-Ⅱ),vascular endothelial growth factor(VEGF),prostacyclin 2(PGI_(2))and thromboconstrictor 2(TXA_(2))were measured by enzyme-linked immunosorbent assay(ELASA);the effect of HSP on coronary artery diastolic function was measured by vascular tension method.Results The TC levels of control group,model group,experimental-L,-M,-H groups were(2.15±0.66),(6.38±1.31),(4.05±0.67),(3.98±1.20)and(3.06±0.92)mmol·L^(-1),respectively;TG levels were(0.89±0.17),(1.44±0.21),(1.45±0.23),(1.15±0.13)and(0.92±0.27)mmol·L^(-1),respectively;LDL-C levels were(0.62±0.12),(1.47±0.37),(1.33±0.35),(1.23±0.36)and(1.01±0.29)mmol·L^(-1),respectively;HDL-C levels were(1.34±0.15),(1.30±0.27),(1.39±0.18),(1.36±0.19)and(1.41±0.11)mmol·L^(-1),respectively;the levels of Ang-Ⅱwere(422.19±12.27),(553.79±13.17),(551.15±11.57),(533.00±11.69)and(496.00±16.67)pg·mL^(-1),respectively;VEGF levels were(184.81±2.45),(234.40±3.15),(226.00±4.87),(209.87±4.94)and(196.59±5.41)pg·mL^(-1),respectively;the levels of PGI2were(67.40±5.76),(86.03±5.02),(86.07±4.15),(70.53±7.03)and(68.59±5.86)pg·mL^(-1),respectively;TXA2levels were(101.44±5.47),(129.41±6.87),(126.50±5.27),(121.31±5.77)and(114.21±8.09)pg·mL^(-1),respectively;th
