摘  要:目的探讨胎盘间充质干细胞(P-MSCs)移植对脓毒症肝损伤小鼠的保护作用。方法将24只雄性C57小鼠随机分为对照组、模型组和实验组,每组8只。对照组小鼠仅开腹游离盲肠后还纳入腹腔,其他2组小鼠用盲肠结扎穿孔法构建脓毒症模型。对照组和模型组均于术后1 h经尾静脉注射0.9%NaCl 0.1 mL;实验组于术后1 h经尾静脉注射P-MSCs 2×105个。术后24 h处死所有小鼠。用酶联免疫吸附试验法检测血清中炎症因子的浓度,用实时荧光聚合酶链反应法检测肝组织中炎症因子基因的表达水平。结果实验组、模型组和对照组血清中肿瘤坏死因子-α含量分别为(568.83±66.39),(776.06±56.14)和(423.85±8.88)pg·mL^(-1),白细胞介素1β(IL-1β)含量分别为(156.24±13.77),(207.88±27.39)和(138.25±19.67)pg·mL^(-1),IL-6含量分别为(181.88±10.42),(214.29±23.49)和(133.65±4.64)pg·mL^(-1),IL-10含量分别为(519.33±28.19),(390.46±11.32)和(321.31±7.70)pg·mL^(-1)。实验组和对照组的上述指标与模型组比较,差异均有统计学意义(均P<0.05)。肝组织炎症因子基因结果趋势与血清一致。结论体外输注的P-MSCs能够显著减轻脓毒症肝损伤小鼠的炎性损伤。Objective To explore the protective effect of placental-derived mesenchymal stem cells(P-MSCs)transplantation on septic liver injury in mice.Methods Twenty-four C57 mice were randomly divided into control group,model group and experimental group,with 8 mice in each group.The mice in the control group were only allowed to open the cecum after laparotomy.The mice in other two groups were treated with cecal ligation and puncture to establish a sepsis model.Mice in the control group and model group were infused with 0.9%NaCl 0.1 mL via tail vein 1 h after operation.Model mice in the experimental group were infused with 2×10~5 P-MSCs via tail vein 1 h after operation.All mice were sacrificed 24 h after surgery.Enzyme-linked immunosorbent assay was used to detect the concentrations of inflammatory factors in serum,quantitative real-time polymerase chain reaction was used to detect the expression levels of inflammatory factor genes in liver tissue.Results The contents of tumor necrosis factorαin the experimental group,model group and control group were(568.83±66.39),(776.06±56.14)and(423.85±8.88)pg·mL^(-1);the contents of interleukin 1β(IL-1β)were(156.24±13.77),(207.88±27.39)and(138.25±19.67)pg·mL^(-1);the contents of IL-6 were(181.88±10.42),(214.29±23.49)and(133.65±4.64)pg·mL^(-1);the contents of IL-10 were(519.33±28.19),(390.46±11.32)and(321.31±7.70)pg·mL^(-1).There were significant differences in the above indicators between the experimental and control groups and the model group(all P<0.05).The trend of inflammatory factor gene results in liver tissue was consistent with that in serum.Conclusion P-MSCs attenuate inflammatory injury in mice with septic liver injury.
