摘  要:目的观察热打击对人脐静脉内皮细胞(HUVEC)P-选择素、E-选择素及细胞间黏附分子-1(ICAM-1)表达的影响。方法将HUVEC细胞分为对照组(细胞始终置于正常细胞培养箱中孵育)、不同温度梯度热打击组(将敷箱温度调整为40℃、41℃、42℃、43℃,细胞置于对应温度培养箱中打击2 h),43℃热打击组(细胞于43℃培养箱中打击2 h,之后放置回正常培养箱中继续培养0 h、3 h、6 h、12 h)。采用Western blot检测HUVEC细胞P-选择素及E-选择素和ICAM-1的表达,采用免疫荧光技术观察P-选择素及E-选择素的分布。结果与对照组比较,热打击组HUVEC细胞E选择素及ICAM-1蛋白表达明显增加,差异具有统计学意义(P<0.05);热打击组复温6 h后E选择素及ICAM-1蛋白的表达均达到峰值,差异均具有统计学意义(P<0.05);热打击组复温12 h后E选择素及ICAMv1蛋白的表达均降到基底水平,差异均具有统计学意义(P<0.05);使用不同温度梯度热打击HUVEC细胞,以及使用43℃热打击HUVEC细胞2 h后复温不同时间点,P选择素的表达均无影响,差异无统计学意义(P>0.05)。结论不同的温度梯度热打击HUVEC细胞对P-选择素的表达均无影响,且热打击后各复温时间点P-选择素的表达亦无差异;但热打击能够明显增加HUVEC细胞E-选择素及ICAM-1蛋白表达。Objective To investigate the effects of heat stress on the expression of P-selectin,E-selectin,and intercellular adhesion molecule-1(ICAM-1)in human umbilical vein endothelial cells(HUVEC).Methods HUVEC cells were divided into control group(cells were incubated in normal cell incubator),gradient heat stress group(cells were incubated at either 40℃,41℃,42℃or 43℃for 2 hours),43℃heat stress group(cells were incubated at 43℃for 2 hours and then in normal cell incubator for 0,3 h,6 h or 12 h).The protein expressions of P-selectin,E-selectin,and ICAM-1 were examined using Western blotting and immunofluorescence confocal microscopy.Results Compared with the control group,the expression levels of E-selectin and ICAM-1 in HUVEC cells increased significantly in heat stress groups(P<0.05),and they reached the peak after rewarming for 6 h(P<0.05),and dropped to the basal level after rewarming for 12 hours(P<0.05).The expression of P-selectin showed no statistically significant differences among gradient heat stress group at different temperature and in 43℃heat stress group with different rewarming time points,P>0.05.Conclusion Heat stress at different temperatures has no effect on the expression of P-selectin in HUVEC cells,and there is no significant difference in the expression of P-selectin among different rewarming time points after 2 hours of heat stress at 43°C.Heat stress can significantly increase the expression of E-selectin and ICAM-1 in HUVEC cells.
