摘 要:为研究黄芪内生枯草芽孢杆菌HS8的抗菌活性及其次生代谢产物,采用琼脂扩散法对黄芪内生枯草芽孢杆菌HS8的抗菌活性进行测定,利用高效液相色谱法(High performance liquid chromatography,HPLC)对黄芪内生枯草芽孢杆菌HS8次生代谢产物进行分析,使用D_(101)大孔吸附树脂柱色谱、聚酰胺柱色谱、硅胶柱色谱及重结晶等方法对黄芪内生枯草芽孢杆菌HS8次生代谢产物进行分离。结果显示,黄芪内生枯草芽孢杆菌HS8发酵液具有较好的抗菌活性,从黄芪内生枯草芽孢杆菌HS8发酵液中分离得到3个化合物,分别为原儿茶酸(A)、β-谷甾醇(B)和阿魏酸(C)。研究结果表明,黄芪内生枯草芽孢杆菌HS8含有大量次生代谢产物,是一株具有深入开发应用价值的内生细菌。To investigate the antibacterial activities and secondary metabolites of endophytic Bacillus subtilis named HS8 which was isolated from the root of Astragalus membranaceus(Fisch.)Bge.,the antibacterial abilities of HS8 were examined by agar diffusion and the secondary metabolites were analyzed by high performance liquid chromatography(HPLC).Meanwhile,the secondary metabolites were isolated and purified by D_(101)macroporous resin,polyamide column chromatography and silica gel column chromatography as well as recrystallization.The results showed that the fermented broth of HS8 had good antibacterial activities and three compounds were separated from the fermented broth of HS8,including protocatechuic acid(A),β-sitosterol(B)and ferulic acid(C).The results indicated that HS8 could produce a lot of active compounds and was an endophytic Bacillus subtilis with practical application value.
