摘  要:目的 基于网络数据库研究王灿辉教授自拟健脾益胃汤治疗慢性萎缩性胃炎的潜在作用机制。方法 检索TCMSP数据库、PubChem数据库、SwissTarget Prediction平台获得健脾益胃汤的作用靶点。检索OMIM、Gene cards数据库,获得慢性萎缩性胃炎的疾病靶点。使用Cytoscape软件构建“药物-化学成分-潜在作用靶点”网络。利用String数据库、Cytoscape软件进行潜在靶点的蛋白互作网络分析,得到主要靶点。通过Metascape平台,对主要靶点进行富集分析。结果 通过检索筛选得到63个主要化学成分,共涉及109个潜在作用靶点,靶点类型25种(包含各种酶、蛋白、受体、离子通道、转录因子等)。对其中主要的45个靶点进行富集分析发现,健脾益胃汤主要通过参与癌症通路、癌症中的蛋白多糖通路、肿瘤坏死因子信号通路、EGFR酪氨酸激酶抑制剂抵抗、AGE-RAGE信号通路、光动力疗法诱导NF-kB存活信号转导、光动力疗法诱导AP-1存活信号、RANKL/RANK(NFKB(配体)受体激活剂)信号通路、氧化应激反应、凋亡信号通路、白细胞分化、炎症反应的调节、对氧气水平的反应、DNA结合转录因子活性的调控、对机械刺激的反应、活性氧代谢过程白介素信号等生物过程发挥对慢性萎缩性胃炎的治疗作用。结论 健脾益胃汤对慢性萎缩性胃炎的治疗作用体现了中药多成分、多靶点、多通路的特点,为阐释其治疗慢性萎缩性胃炎的作用机制提供了科学依据,也为今后新药研发提供了参考。Objective: To study the potential mechanism of decoction for strengthening spleen and tonifying stomach(Jianpi Yiwei Decoction) created by Professor Wang Canhui in the treatment of chronic atrophic gastritis based on network database. Methods: The target of Jianpi Yiwei Decoction was obtained by searching TCMSP database, PubChem database and SwissTarget Prediction platform. The disease targets of chronic atrophic gastritis were obtained by searching OMIM and Gene cards databases. Cytoscape software was used to establish the network of Drug-Chemical Component-Potential Targets. By using Cytoscape software and String database, the protein interaction network of potential targets was analyzed to obtain the main targets. The main targets were made enrichment analysis through Metascape platform. Results: 63 main chemical components were obtained through retrieval and screening, involving 109 potential targets, 25 types of targets(including various enzymes, proteins, receptors, ion channels, transcription factors, etc.). The enrichment analysis of 45 major targets showed Jianpi Yiwei Decoction mainly played a therapeutic role in chronic atrophy gastritis by participating in cancer pathway, protein polysaccharide pathway in cancer, tumor necrosis factor signaling pathway, EGFR tyrosine kinase inhibitor resistance, AGE-RAGE signaling pathway, NF-kB survival signal transduction induced by photodynamic therapy, AP-1survival signal induced by photodynamic therapy, RANKL/RANK(NFKB)(ligand) receptor activator signaling pathway, oxidative stress response, apoptotic signaling pathway, leukocyte differentiation, regulation of inflammatory response, response to oxygen level, regulation of DNA-binding transcription factor activity, response to mechanical stimulation, active oxygen metabolism process interleukin signaling and other biological processes. Conclusion: The therapeutic effect of Jianpi Yiwei Decoction on chronic atrophic gastritis reflects the characteristics of multi-component, multi-target and multi-pathway of traditi
