低pH值环境乙酸H_(2)O_(2)磺胺甲基嘧啶协同酮康唑抑制白色假酵母菌生长

摘  要:目的 对低pH值环境乙酸H_(2)O_(2)磺胺甲基嘧啶协同酮康唑抑制白色假酵母菌生长情况进行研究,为外阴阴道假丝酵母菌病发病原因及治疗提供依据。方法 制备pH值为3~6培养肉汤、含5.0%~0.1%乙酸培养肉汤及含0.3%~0.03%H_(2)O_(2)培养肉汤,配制并使pH值为3、6的培养肉汤及上述含乙酸、H_(2)O_(2)培养肉汤中含250.000μg/ml~0.061μg/ml酮康唑,调肉汤培养基含酮康唑、磺胺甲基嘧啶浓度为2 000.000μg/ml~0.122μg/ml,肉汤含酮康唑:磺胺甲基嘧啶浓度为2 000.000μg/ml:0.008μg/ml~0.122μg/ml:125.000μg/ml,白色假丝酵母菌经上述肉汤培养48 h,观察并检测其生长抑制情况。结果 pH值为3、4、6时的生长抑制率分别为65.21%、63.54%和1.96%,乙酸H_(2)O_(2)浓度依赖性抑制白色假丝酵母菌生长,pH值为3能增加酮康唑抑制白色假丝酵母菌生长能力,而乙酸和H2O与酮康唑间协同抑制生长能力较弱。与磺胺甲基嘧啶联合,酮康唑最低抑菌生长浓度降低16倍。结论 低pH值环境乙酸H_(2)O_(2)能抑制白色假丝酵母菌生长,pH值环境、磺胺甲基嘧啶协同,能增加酮康唑抑菌生长能力;乙酸H_(2)O_(2)等浓度明显下降,减弱甚至失去抑菌生长能力也与VVC发生有关。Objective This paper aims to evaluate the suppressing effect of ketoconazole combined with pH acetic acid H_(2)O_(2)sulfamethazine on Candida albicans growth in order to provide evidence for the pathogenesis and treatment of Vulvovaginal Candidiasis(VVC). Methods Culture broth with pH 3-6 and the culture broth with 5.0%-0.1% acetic acid and 0.3%-0.03% H_(2)O_(2)were prepared. Culture broth with pH 3 and pH 6 and the culture broth with 5.0% acetic acid, 0.5% acetic acid, and 0.3% H_(2)O_(2)and 0.03% H_(2)O_(2)were chosen, and 250.000 μg/ml-0.061 μg/ml ketoconazole was added to the above broth. 2 000.000 μg/ml-0.061 μg/ml ketoconazole mixed with 0.008 μg/ml-125.000 μg/ml sulfamethazine was prepared and added to broth. Candida albicans were cultured in above broth for 48 h. The growth inhibition of Candida albicans was detected. Results Growth suppression rate was 65.21%, 63.54% and 1.96% when pH was 3, 4 and 6. Growth and biofilm were suppressed by pH lactic acid acetic acid H_(2)O_(2)in concentration-dependent manner. The Suppression of ketoconazole was increased significantly when pH was 3. The synergistic effect was weaker between acetic acid H_(2)O_(2)and ketoconazole. The MIC of ketoconazole combined with sulfamethazine was reduced by 16 times. Conclusion Candida albicans growth can be suppressed by low pH acetic acid and H_(2)O_(2). The inhibition of ketoconazole was increased significantly when it combined with low pH environment and sulfamethazine. VVC may be related to the decrease or loss of suppression on Candida albicans growth duo to the decrease of acetic acid and H_(2)O_(2)concentrations.
