摘 要:猪非典型瘟病毒(atypical porcine pestivirus,APPV)是引起仔猪先天性震颤(congenital tremor,CT)的主要病原,给养猪业带来严重的经济损失。为了解APPV在广东地区猪群的流行和变异情况,本研究于2018—2019年从广东地区采集12份疑似CT的病料样品,通过RT-PCR对APPV进行检测,并对APPV阳性的病料样品通过PCR扩增其全长基因组序列并测序,同时对其ORF和E2基因的同源性、遗传进化关系及E2蛋白氨基酸分子特征进行分析。结果显示,有3份样品为APPV阳性,进一步对其中1份阳性样品通过全基因组PCR扩增得到1条长度为10989 bp的APPV全长序列,命名为APPV-China/GD-ZK/2018(GD-ZK)。GD-ZK与参考毒株ORF和E2基因的核苷酸同源性分别为80.7%~99.7%,80.8%~99.9%,其中与APPV/CNYJ/2018的同源性最高。分别对GD-ZK株的ORF和E2基因进行遗传演化分析,结果显示,根据遗传进化关系可将APPV划分为Genotype 1、Genotype 2和Genotype 3三种基因型,而本研究中鉴定到的GD-ZK株属于Genotype 2,与APPV/CNYJ/2018的进化关系最近。分子特征分析结果显示GD-ZK株E2蛋白发生了S^(54)T和S^(130)T 2个特征性的氨基酸位点突变,且该突变均存在于Genotype 2。因此这2个氨基酸位点突变可作为APPV基因分型潜在的分子标记。本研究初步揭示了APPV在广东地区猪群中的遗传变异情况,为掌握APPV在广东地区的流行本底奠定了基础。Atypical porcine pestivirus(APPV)is a major pathogen causing congenital tremor(CT)in piglets,causing serious economic losses to the pig industry.To address the evolutionary and variation of APPV in swine in Guangdong Province,12 samples of suspected CT were collected from Guangdong Province in this study during 2018 to 2019.APPV detection was performed by RT-PCR,and full-length genome of APPV was amplification for samples that were positive for APPV.Furthermore,the evolutionary and molecular characterization analysis for ORF and E2 gene of APPV was also conducted.The results showed that three samples were positive for APPV identified by RT-PCR tests.Subsequently,a nearly full-length genome of APPV with 10989 bp in length,named APPV-China/GD-ZK/2018(GD-ZK),was obtained by PCR amplification and sequencing.The nucleotide homology of ORF and E2 gene between GD-ZK and other reference strains ranged from 80.8%to 99.7%and 80.8%to 99.9%,respectively,with the highest identification with APPV/CNYJ/2018.The evolutionary analysis of ORF and E2 gene for GD-ZK was conducted,respectively,and the results showed that APPV could be classified into three genotypes,genotype 1,genotype 2 and genotype 3.GD-ZK strain identified in this study belonged to genotype 2 and cluster with APPV/CNYJ/2018 based on evolutionary relationship.The molecular characterization analysis showed that two amino acids mutation(S^(54)T and S^(130)T)were identified in E2 protein of GDZK,which was also present in strains belongs to genotype 2,and these mutations could be used as potential molecular markers for APPV genotyping.The epidemiology and evolutionary analysis of APPV was conducted in Guangdong Province in this study,which is helpful to know about the epidemiology and prevention,as well as the prevention of APPV in Guangdong Province.
