摘  要:采用酸溶法提取鹿茸根部骨化组织中可溶性钙,探究不同酸种类、酸用量、提取时间、提取温度对鹿茸根部骨化组织可溶性钙含量的影响,通过正交试验优化骨钙提取的最佳工艺条件,检验不同浓度骨钙对Caco-2细胞的毒性作用,并构建Caco-2模型模拟体内肠道吸收环境,研究骨钙的转运效率。结果表明,最佳提取条件即苹果酸提取,酸用量为5%,提取温度为55℃,提取时间为70 min时,骨钙含量为(6.136±0.015)mg/g。体外模拟结果表明,骨钙对Caco-2细胞呈现低毒性作用且转运效率为CaCl2的2倍左右。该研究为开发新的钙源和提高鹿茸根部骨化组织的附加值提供了新的发展方向。The soluble calcium was extracted from antler bone by acid-soluble method,and the effects of different acid types,acid concentration,extraction time and extraction temperature on the soluble calcium content of antler bone were investigated.The optimal process conditions of bone calcium extraction were determined by orthogonal tests.The effect of bone calcium on Caco-2 cytotoxicity was determined,and the Caco-2 model was established to simulate the intestinal absorption environment for studying the transport efficiency of bone calcium.The results showed that the optimal extraction process was:extrated by malic acid,malic acid dosage of 5%,extraction temperature of 55℃and extraction time of 70 min.The calcium content of antler bone was(6.136±0.015)mg/g under the optimal process conditions.The in vitro simulation results showed that antler bone calcium showed low toxicity in Caco-2 cells and the transport efficiency was about two times that of CaCl2.This study provides a new development direction for developing new calcium sources and improving the added value of antler root ossification tissue.
