摘 要:为探究幼穗盐胁迫下上调表达的OsMYB家族基因的生物学特性及其在响应盐胁迫中的功能,本研究利用水稻幼穗在不同时间盐胁迫处理的转录组数据,筛选到了17个表达上调的OsMYB基因。生物信息学分析结果表明:17个OsMYB分别位于9条水稻染色体上,且大部分OsMYB基因有1~2个内含子结构,共鉴定到了8个motif。motif 1基序存在于17个OsMYB蛋白中。15个OsMYB蛋白包含motif 2和motif 3基序,且motif 7仅存于6个OsMYB蛋白中。在17个OsMYB基因启动子区中鉴定了多个非生物胁迫响应元件,推测这17个OsMYB基因成员可能参与了植物响应某些胁迫的生物学过程,并且在植物胁迫响应过程中发挥了重要作用。表达分析结果表明:MH01g0044200、MH07g0422700、MH07g0545300这3个MYB的表达水平随着NaCl处理时间而上升,其余的14个OsMYB都在NaCl处理6 h后表达水平达到最高。MH01g0227400、MH04g0517900、MH05g0402100、MH05g0514400这几个MYB在NaCl处理24 h后的表达水平要明显低于处理0 h。这表明17个水稻MYB基因可能在水稻幼穗响应盐胁迫过程中行使重要且不同的功能。通过比对58M和58L中17个MYB基因的表达情况,最后筛选出1个关键基因MH12g0086600做后续研究,为下一步开展水稻幼穗盐胁迫响应的分子机制研究提供了参考依据。In order to explore the biological characteristics of the up-regulated OsMYB family genes under salt stress in young panicles and their functions in response to salt stress,this study used the transcriptome data of rice young panicles treated with salt stress at different times to screen 17 up-regulated OsMYB genes.The results of bioinformatics analysis showed that 17 OsMYBs were located on 9 rice chromosomes,and most of the OsMYB genes had 1~2 intron structures.A total of 8 motifs were identified.The motif 1 motif is present in 17 OsMYB proteins.Fifteen OsMYB proteins contained motif 2 and motif 3 motifs,and motif 7 was present in only 6 OsMYB proteins.Multiple abiotic stress response elements were identified in the promoter regions of 17 OsMYB genes.It is speculated that these 17 OsMYB gene members may be involved in the biological process of plants responding to certain stresses and play an important role in the process of plant stress response.The expression analysis results showed that the expression levels of three MYBs,MH01g0044200,MH07g0422700,and MH07g0545300,increased with the time of Na Cl treatment,and the other 14 OsMYBs all reached their highest expression levels after6 h of Na Cl treatment.The expression levels of MYBsMH01g0227400,MH04g0517900,MH05g0402100 and MH05g0514400 after Na Cl treatment for 24 h were significantly lower than those treated for 0 h.This indicated that 17 rice MYB genes may play important and different functions in the response of rice young panicles to salt stress.By comparing the expression of 17 MYB genes in 58M and 58L,a key gene MH12g0086600 was finally screened for follow-up research,which provided a reference for the next step to carry out research on the molecular mechanism of rice young panicle response to salt stress.
