摘  要:AMEP是来自于枯草芽孢杆菌的一种新型蛋白激发子,可诱导植物叶片发生过敏反应并激发植物的防卫反应,提高植物的抗病性,是生物农药的理想候选。为了研究AMEP蛋白的聚合体状态与金属离子之间的关系,以及蛋白多聚体对其活性的影响,比较了不同浓度Fe^(2+)对AMEP蛋白激发子的聚合程度和活性的影响,设置0.03、0.3、3 g·L^(-1)Fe^(2+)3种浓度梯度的Fe^(2+)与AMEP蛋白混合溶液处理烟草叶片,并通过过敏反应、酶活、灰霉抗病试验对烟草叶片的生理指标进行检测。低浓度Fe^(2+)能够增加AMEP聚合程度提高蛋白活性,从而增强烟草的过敏反应和早期防御反应和某些防御酶的激发,从而增加对灰霉病的抗性;而高浓度Fe^(2+)则会降低蛋白活性。综上,低浓度的Fe^(2+)与AMEP蛋白结合形成了更多适宜其发挥蛋白性能的多聚体,能更好地保存酶解位点以便其发挥其性能,从而提高AMEP蛋白的活性。AMEP is a new Protein Elicitor from Bacillus subtilis,which can induce hypersensitivity reaction in plant leaves,stimulate plant defense reaction and improve plant disease resistance.It is an ideal candidate for biological pesticides.In order to investigate the relationship between the polymeric state of AMEP protein and metal ions,and the effect of protein poly on its activity,this experiment compared the influence of different concentrations of Fe^(2+)on the polymerization degree and activity of AMEP protein activators.Tobacco leaves were treated with mixed solution of Fe^(2+)and AMEP protein in three concentration gradients of 0.03 g·L^(-1)Fe^(2+),0.3 g·L^(-1) Fe^(2+)and 3 g·L^(-1)Fe^(2+).The physiological indexes of tobacco leaves were detected by allergic reaction,enzyme activity and gray mold resistance test.Low concentration of Fe^(2+)could increase the polymerization degree of AMEP and protein activity,thus enhancing the allergic reaction and early defense reaction of tobacco and the stimulation of some defense enzymes,thus increasing the resistance to gray mold.However,high concentration of Fe^(2+)decreased the protein activity.In conclusion,low concentration of Fe^(2+)combines with AMEP protein to form more suitable polymers,which not only can better preserve the hydrolysis sites for its performance,but also improve the activity of AMEP protein.
