摘  要:试验旨在研究添加不同种类、浓度抗氧化剂的精液稀释液对液态保存的鸡精液质量和受精能力的影响。试验以Lake's液为基础稀释液,分别加入0.5mmol/LN-乙酰半胱氨酸(T1组)、1mmol/LN-乙酰半胱氨酸(T2组)、0.5mmol/LL-半胱氨酸(T3组)、1mmol/LL-半胱氨酸(T4组)、1ng/L硒+8ng/L维生素E(T5组),另设空白对照组(T6组),每组3个重复;然后采集公鸡精液,按1:2倍比分别将6组稀释液加入并混匀,5℃下冷藏保存0、4、12、24h后进行显微镜观察,并取保存4h后的精液进行人工授精和种蛋孵化。结果显示:精子活力随着储存时间的增加显著降低(P<0.05),其中4、12、24h的T5组精子活力最高且在12、24h显著高于T1、T3、T6组(P<0.05);T5组精子质膜完整率显著高于T1、T3、T6组(P<0.05),而丙二醛含量相反;T5组受精率显著高于T1、T6组(P<0.05),T2、T4组受精率均显著高于T1组(P<0.05);各组间孵化率无显著差异(P>0.05)。研究表明,Lake's液中添加1ng/L硒+8ng/L维生素E有助于提高精子活力、保护精子质膜完整率并降低氧化应激反应,从而提高受精率和孵化率。This study was aimed to analyze the effects of semen extender which added antioxidants with different kinds or concentrations on quality and fertilizing ability of chicken semen in liquid storage. As a basic extender, Lake’s was added with0.5 mmol/L N-acetyl-cysteine(group T1), 1 mmol/L N-acetyl-cysteine(group T2), 0.5 mmol/L L-cysteine(group T3), 1 mmol/L L-cysteine(group T4) and 1 ng/L Se+8 ng/L vitamin E(group T5) and blank control group(group T6). The fresh semen was diluted to 2 times by semen extender, then mixed well. The samples were preserved in 5 ℃, which was examined in microscope after0, 4, 12, 24 h and was used for artificial in semination and incubation after 4 h, respectively. The results showed that sperm motility decreased significantly with the increase of storage time(P<0.05). The sperm motility of group T5 which stored for 4 h, 12 h and24 h was the highest and significantly higher than that of group T1, T3 and T6 for 12 h and 24 h(P<0.05). The plasma membrane integrity of chicken sperm in group T5 was significantly higher than that in group T1, T3 and T6(P<0.05), while the content of malondialdehyde was opposite. The fertilizing rate of group T5 was significantly higher than that of group T1, T6(P<0.05), while the fertilizing rate of group T2 and T4 were significantly higher than that of group T1(P<0.05). The hatching rate among groups had no significant differences(P>0.05). It’s concluded that Lake’s +1 ng/L Se+8 ng/L VEcould effectively improve sperm motility, protect sperm plasma membrane integrity, reduce sperm oxidative stress to increase fertilizing rate and hatching rate.
