摘  要:为筛选适合传染性法氏囊病病毒(IBDV)增殖的细胞系,试验对IBDV Ts株感染DF-1细胞与Vero细胞的致细胞病变效应(CPE)、病毒粒子密度和病毒基因组ds RNA含量差异、半数组织感染量(TCID_(50))和VP2基因表达水平等进行检测,并绘制IBDV Ts株在两种细胞中的一步生长曲线。结果显示:在相同感染复数的IBDV感染后,DF-1细胞CPE程度高于Vero细胞;DF-1细胞中IBDV病毒粒子密度和ds RNA含量均高于Vero细胞;IBDV感染DF-1细胞冻融采集的上清液中病毒滴度(TCID_(50)=10^(6.500±0.057)/0.1 mL)显著高于Vero细胞(TCID_(50)=10^(5.645±0.113)/0.1 mL),VP2基因表达量显著高于Vero细胞(P<0.05);一步生长曲线显示IBDV Ts株在DF-1细胞中达到最大病毒滴度的时间(60 h)早于Vero细胞(72 h),且DF-1细胞中最大病毒滴度(TCID_(50)=10^(8.250±0.204)/0.1 mL)高于Vero细胞中(TCID_(50)=10^(7.417±0.118)/0.1 mL)。研究表明,DF-1细胞比Vero细胞更适合作为IBDV Ts株增殖和致病机理研究的工具细胞。In order to screen out suitable cell lines for infectious bursal disease virus (IBDV) proliferation,the cytopathic effect (CPE),virion density and viral genomic dsRNA content,the median tissue culture infective dose (TCID50) and VP2 gene expression were compared between DF-1 cells and Vero cells infected with Ts strain of IBDV.The results showed that,after infection with the same multiplicity of Ts strain of IBDV,the degree of CPE in DF-1 cells was stronger than that in Vero cells;Virion density and viral genomic ds RNA content in DF-1 cells were more than those in Vero cells;The supernatant of DF-1 cells and Vero cells infected with Ts strain of IBDV by freezing and thawing were collected,and the viral titer in the collected supernatant from IBDV-infected DF-1 cells (TCID_(50)=10^(6.500±0.057)/0.1 mL) was significantly higher than that from IBDV-infected Vero cells(TCID_(50)=10^(5.645±0.113)/0.1 mL);The expression of VP2 gene in IBDV-infected DF-1 cells was higher than that in Vero cells (P<0.05);The one-step growth curve showed that Ts strain reached the maximum virus titer in DF-1 cells at 60 h,which was earlier than that in Vero cells (72 h),and the maximum virus titer in DF-1 cells (TCID_(50)=10^(8.250±0.204)/0.1 mL) was higher than that in Vero cells(TCID_(50)=10^(7.417±0.118)/0.1 mL).These results suggested that DF-1 cells were more suitable for the study of propagation and pathogenesis of Ts strain of IBDV than Vero cells.
