摘  要:为了解猪脾转移因子(transfer factor,TF)对新城疫病毒(Newcastle disease virus,NDV)La Sota弱毒疫苗株免疫效果的影响,本研究采用10^(5.17)EID_(50)La Sota株单独免疫(单独免疫1组)或与TF联合免疫(联合免疫1组)SPF鸡,14 d后以10^(4.7)ELD_(50)F_(48)E_(9)株首次攻毒,设立对照1组(非免疫/首次攻毒),首次攻毒14 d后以10^(9.1)ELD^(50)F_(48)E_(9)株二次攻毒,设立对照2组(非免疫/非首次攻毒/二次攻毒)和空白1组(非免疫/非攻毒),通过淋巴细胞增殖率测定法、血凝抑制试验、荧光定量RT-PCR方法检测T淋巴细胞转化水平(PHA-P SI)、B淋巴细胞转化水平(LPS SI)、新城疫(ND)抗体效价、病毒血症、排毒与主要组织的F48E9株感染情况。结果显示:首次攻毒后,单独和联合免疫1组保护率均为100%,对照1组病死率为100%;二次攻毒后,单独和联合免疫1组保护率分别为20%和50%、病死率分别为60%和40%,对照2组病死率为100%。免疫后14 d,联合免疫1组的PHA-P SI、LPS SI和抗体效价均极显著高于单独免疫1组(P<0.01)。首次攻毒后7~14 d,联合免疫1组的PHA-P SI、LPS SI和抗体效价均极显著高于单独免疫1组(P<0.01),首次攻毒后单独和联合免疫1组均没有检测出病毒血症、泄殖腔和口咽排毒。二次攻毒后3~7 d,联合免疫1组的PHA-P SI、LPS SI和抗体效价均极显著高于单独免疫1组(P<0.01),二次攻毒后联合免疫1组鸡病毒血症率、病毒血症时间、鸡泄殖腔和口咽排毒时间、排毒鸡比例、鸡肝、脾、肺、肾、法氏囊、盲肠扁桃体、脑等主要组织NDV F_(48)E_(9)株感染阳性率均明显低于单独免疫1组。结果表明,TF可提高La Sota株攻毒保护率,提高La Sota株、NDV F_(48)E_(9)株接种后鸡T、B淋巴细胞转化水平和ND抗体效价,降低攻毒后病毒血症率、病毒血症时间、鸡排毒时间、排毒鸡比例和NDV F_(48)E_(9)株在鸡体内的复制。The aim of this study is to evaluate the effect of pig spleen transfer factor(TF)on the immune efficacy of Newcastle disease virus(NDV)attenuated vaccine La Sota strain.Specific pathogen free chickens were inoculated with 10^(5.17)EID_(50)NDV strain La Sota singlely(single vaccine group 1),NDV strain La Sota combined with TF(co-vaccination group 1),respectively,and challenged with 10^(4.7)ELD_(50)virulent NDV strain F_(48)E_(9)on 14 d post vaccination(dpv)and 10^(9.1)ELD_(50)virulent NDV strain F_(48)E_(9)on 14 d post the first challenge(dpfc).The control group 1(nonimmunized but the first challenged),control group 2(non-immunized and non-the first challenged but the second challenged)and blank group 1(non-immunized and non-challenged)were set up simultaneously.In addition,the T lymphocyte transformation level(PHA-P SI),B lymphocyte transformation level(LPS SI),Newcastle disease(ND)antibody titer,viremia,virus shedding and infection of F_(48)E_(9)strain in main tissues were detected by lymphocyte proliferation assay,hemagglutination inhibition test and fluorescence quantitative RT-PCR.The results showed that after the first challenge,the protection rate of single vaccine group 1 and co-vaccination group 1 was 100% and the mortality of control group 1 was 100%.After the second challenge,the protection rates of single vaccine group 1 and co-vaccination group 1 were 20% and 50%,respectively,and their mortality was 60% and 40%.In the meantime,the mortality of control group 2 was 100%.On 14 dpv,the PHA-P SI,LPS SI and antibody titer in co-vaccination groupl were significantly higher than those in single vaccine group 1(P<0.01).From 7 dpfc to 14 dpfc,the PHA-P SI,LPS SI and antibody titer in co-vaccination group 1 were significantly higher than those in single vaccine group 1(P<0.01).In addition,after the first challenge,the viremia,virus shedding from cloacal and oropharyngeal could not be detected in single vaccine group 1 and co-vaccination group 1.From 3 d post the second challenge(dpsc)to 7 dpsc,the PHA-P SI,LPS S
