摘　　要：为了探讨绵羊microRNAs(miRNAs)是否靶向TNF 3’UTR区域,以及初步确定miRNAs靶向其基因的区域,笔者以绵羊miRNAs为研究对象,利用PCR方法克隆TNF 3’UTR(3’非翻译区),应用miRanda软件分析miRNAs靶向TNF 3’UTR基因的具体区域和结合能力。结果表明,成功克隆TNF 3’UTR,大小为500 bp;miR-125b、let-7b和let-7c同时靶向TNF 3’UTR基因,表明宿主miRNAs具有潜在的靶向抑制TNF 3’UTR基因表达的能力,初步明确miRNAs靶向TNF 3’UTR基因的具体区域,为进一步探讨miRNAs调节TNF 3’UTR表达提供了基础资料。In order to investigate whether sheep microRNAs(miRNAs)target the TNF 3’-untranslated region(3’UTR),and to preliminarily determine the region of miRNAs targeting their genes,sheep miRNAs were used as the research object;TNF 3’UTR was cloned by PCR method,and miRNAs targeting TNF 3’UTR and binding capacity to the TNF 3’UTR gene were analyzed by miRanda software.The results showed that TNF 3’UTR was successfully cloned with a size of 500 bp;miR-125b,let-7b and let-7c simultaneously targeted the TNF 3’UTR gene,indicating that host miRNAs have potential targets to inhibit the expression of TNF 3’UTR gene.The ability of miRNAs to target the specific region of TNF 3’UTR gene is preliminarily identified,which provides basic data for further research on the regulation of TNF 3’UTR expression by miRNAs.
