摘 要:目的 研究肉苁蓉复方对慢性疲劳综合征模型小鼠的药效作用。方法 通过力竭、束缚和冷水刺激等建立小鼠慢性疲劳综合症模型,用力竭游泳时间、悬尾和旷场实验对小鼠行为进行测定;测定小鼠血清中乳酸、尿素氮和肝/肌糖原含量;透射电镜观察小鼠骨骼肌线粒体形态。结果 与空白组小鼠相比,模型组力竭游泳时间和旷场活动箱内中央路程减少,肝/肌糖原降低,累计不动时间延长,尿素氮和乳酸含量增加,差异有统计学意义(P<0.05)。与模型组小鼠相比,给药组力竭游泳时间和旷场活动箱内中央路程延长,肝/肌糖原增加,累计不动时间缩短,尿素氮和乳酸含量降低,差异有统计学意义(P<0.05);给药组骨骼肌线粒体膜结构完整,形态接近空白组。结论 肉苁蓉复方能改善小鼠行为,改善骨骼肌肌丝疏松程度,增加线粒体数目和修复结构受损线粒体,对慢性疲劳综合征小鼠有抗疲劳的作用。Objective To study the efficacy of Cistanche deserticola compound on chronic fatigue syndrome model mice. Methods The chronic fatigue syndrome model of mice was established by exhaustion, restraint and cold water stimulation. The behavior of mice was measured by exhaustive swimming time, tail suspension and open field test.The contents of lactic acid, urea nitrogen and liver/muscle glycogen in serum of mice were determined. Mitochondrial morphology of skeletal muscle in micewasobserved by transmission electron microscope. Results Compared with the blank group, the exhaustive swimming time and the central distance in the open field activity box of the model group was decreased;the liver/muscle glycogen wasdecreased;the cumulative immobility time wasprolonged;the urea nitrogen and lactic acid content wasincreased, and the difference was statistically significant(P<0. 05). Compared with the model group, the exhaustive swimming time and the central distance in the open field activity box were prolonged;the liver/muscle glycogen was increased;the cumulative immobility time was shortened;and the urea nitrogen and lactic acid contents were decreased in the administration group(P<0. 05). The mitochondrial membrane structure of skeletal muscle in the administration group was complete;and the morphology was close to the blank group. Conclusion Cistanche deserticola compound can improve the behavior of mice, and it improve the degree of loose skeletal muscle filaments, and it couldincrease the number of mitochondria and repair damaged mitochondria, and it has anti-fatigue effect on chronic fatigue syndrome mice.
