摘  要:为了解山东省枣庄市2021-2022监测年度乙型流感病毒Victoria系的流行情况,分析病毒全基因组遗传特征,了解病毒进化变异特点,分析与疫苗株的相似程度,为流感的防控提供参考依据。从枣庄市流感监测网络实验室分离的流感毒株中随机选取10株B/Victoria系毒株,以WHO推荐的B/Victoria流感疫苗株B/Washington/02/2019(2020-2022北半球疫苗代表株)为参考进行全基因序列测定,利用生物信息学软件构建进化树,并分析分子进化特征。2021年4月-2022年3月,枣庄市流感病毒监测阳性率为17.03%(202/1186),均为B/Victoria系流感。将枣庄市分离的10株B/Victoria系毒株序列与北半球疫苗代表株B/Washington/02/2019相比血凝素(Hemagglutinin,HA)基因核苷酸同源性为99.5%~100%,HA基因氨基酸同源性为99.7%~100%;HA进化树分析中,该10株B/Victoria系毒株与疫苗代表株B/Washington/02/2019同属于1A分支;但发现已有5处抗原表位突变,均发生在120-loop区122位、133位,150-loop区144位、150位,190-loop区197位;神经氨酸酶(NA)未发现关键氨基酸位点变异。NA抑制剂耐药位点均未发生变异,奥司他韦等药物治疗流感敏感。枣庄市2021-2022监测年度流行的流感病毒均为乙型Victoria系,与疫苗推荐株相比HA发生了抗原漂移,提示B/Victoria系疫苗株与本地区流行株的匹配度有所降低。枣庄地区流行的乙型流感病毒对药物奥司他韦和扎那米韦依然敏感。今后继续开展流感病毒分子进化特征监测是重要的,有利于了解乙型流感流行风险,以及对流感疫苗和治疗药物效果的影响。To analyze the epidemiological characteristics and the whole genome characteristics of the Victoria lineage of influenza B virus in 2021 surveillance in Zaozhuang, Shandong Province, China;to understand the evolutionary variation characteristics and match to the vaccine strain;and to Provide a reference for the prevention and control of influenza. Ten B/Victoria lineage strains were randomly selected from influenza strains isolated from the surveillance network laboratory in Zaozhuang. The full gene sequence was determined with reference to the influenza vaccine strain B/Washington/02/2019(representative strain of the Northern Hemisphere vaccine 2020-2022 recommended by the WHO). An evolutionary tree was constructed by bioinformatics software and the characteristics of molecular evolution were analyzed. The positive rate of influenza virus was 17.03%(202/1,186) in Zaozhuang from April 2021 to March 2022, and all isolates were B/Victoria strains. Compared with influenza vaccine strain B/Washington/02/2019, the nucleotide homology of the hemagglutinin(HA) gene was 99.5%, the amino acid homology of the HA gene was 99.5%-100%, and the amino acid homology of the HA gene was 99.7%-100%. In the evolutionary tree analysis of the HA gene, the 10 B/Victoria strains and influenza vaccine B/Washington/02/2019 strains belonged to the 1A branch.However, five antigenic epitope mutations were found, all occurring at 122 and 133 in the 120-loop region, 144and150 in the 150-loop region, and 197 in the 190-loop region;however, no variation in key amino acid sites of neuraminidase(NA) were observed. The influenza viruses prevalent in Zaozhuang,China from 2021 to 2022were all type B Victoria strains, and HA had antigen drift compared with the recommended vaccine strain. It is suggested that the degree of matching between B/Victoria vaccine strains and the epidemic strain in this area has decreased. Influenza B virus prevalent in the Zaozhuang area is still sensitive to Oseltamivir and Zanamivir. It is important to continue to monitor
