摘 要:目的探究腹腔感染小鼠血管内皮细胞损伤相关细胞因子随时间的变化规律。方法盲肠结扎穿孔手术构建腹腔感染小鼠模型,并将小鼠分为实验组和对照组。通过Luminex多细胞因子检测两组小鼠0、6、12、24、48及72 h外周血中上皮细胞钙黏蛋白(E-Cadherin)、血管内皮细胞钙黏连蛋白(VE-Cadherin)、细胞间黏附分子-1(ICAM-1)及血管细胞黏附分子-1(VCAM-1)分泌变化情况。结果实验组小鼠在处理后所有检测时点ICAM-1水平均高于对照组(P<0.05)。与0 h基线水平对比,实验组与时间的交互作用在实验组处理后6 h(β=2173.5,P<0.001)、12 h(β=6852.7,P<0.001)、24 h(β=9202.5,P<0.001)、48 h(β=1498.9,P<0.001)及72 h(β=1201.2,P<0.001)差异均有统计学意义。VCAM-1水平在两组小鼠间经处理后的6 h、12 h及48 h差异有统计学意义(P<0.05),实验组小鼠在处理后6 h(β=12640.3,P<0.001)、12 h(β=9156.6,P<0.001)均高于0 h。结论内皮细胞损伤相关因子ICAM-1和VCAM-1可以作为判断小鼠腹腔感染早期的生物标志物,而VE-Cadherin和E-Cadherin则可能与感染后期内皮细胞的破坏相关。Objective To investigate the time-dependent changes of cytokines related to vascular endothelial cells injury in intraperitoneally infected mice.Methods The abdominal infection model was established by cecal ligation and puncture(CLP),and the mice were divided into the experimental group and the control group.The secretion of epithelium-Ca2+dependent cell adhesion molecule(E-Cadherin),vascular endothelial-Ca2+dependent cell adhesion molecule(VE-Cadherin),intercellular adhesion molecule-1(ICAM-1)and vascular cell adhesion molecule-1(VCAM-1)in mice peripheral blood of two groups at 0,6,12,24,48 and 72 h were detected by Luminex multi-cytokine.Results The ICAM-1 values of mice in the experimental group were higher than those in the control group at all detection time points after treatment(P<0.05).Compared with the baseline level at 0 h,the interaction between the experimental group and time was statistically significant at 6 h(β=2173.5,P<0.001),12 h(β=6852.7,P<0.001),and 24 h(β=9202.5,P<0.001),especially at 48 h(β=1498.9,P<0.001),72 h(β=1201.2,P<0.001)after the treatment in the experimental group.There were statistically significant differences in the measured value of VCAM-1 between the two groups of mice at 6,12,and 48 h after treatment(P<0.05).The measured values of VCAM-1 in the experimental group at 6 h(β=12640.3,P<0.001)and 12 h(β=9156.6,P<0.001)after treatment were higher than those at 0 h(the baseline level).Conclusions Endothelial cell related factors ICAM-1 and VCAM-1 can be used as biomarkers to judge the early stage of infection in mice,while VE-Cadherin and E-Cadherin may be related to the destruction of endothelial cells at the later stage of infection.
