摘 要:目的观察糖尿病兔心房炎症和心房纤维化改变及5脂氧合酶(ALOX5)、12脂氧合酶(ALOX12)、单核细胞趋化蛋白1(MCP-1)在心房组织中表达变化。方法选择健康雄性日本大耳白兔13只,随机选取8只白兔建立1型糖尿病兔模型,其中5只建模成功作为糖尿病组,另外5只白兔作为对照组。检查超声心动图、病理指标,并提取左心房组织蛋白进行Western blot检测。结果与对照组比较,糖尿病组室间隔厚度明显增加,LVEF明显降低,差异有统计学意义(P<0.05)。糖尿病组左心房组织胶原容积分数较对照组明显增加[(7.97±1.01)%vs(3.11±0.99)%,P<0.01]。糖尿病组左心房组织ALOX5、ALOX12表达明显高于对照组,差异有统计学意义(P<0.05);2组MCP-1表达比较,差异无统计学意义(P>0.05)。结论ALOX5、ALOX12表达可能与糖尿病兔心房炎症及纤维化相关。Objective To observe the changes of atrial inflammation and atrial fibrosis and explore the expression changes of 5-lipoxygenase(ALOX5),12-lipoxygenase(ALOX12)and monocyte chemoattractant protein 1(MCP-1)in the atrium of diabetic rabbits.Methods Thirteen healthy male Japanese white rabbits were subjected,and 8 of them were randomly selected to establish a rabbit model of type 1 diabetes mellitus(T1 DM).Finally,5 rabbits with successful modeling were assigned into the diabetes group,and the other 5 rabbits into the control group.Echocardiography and pathological observation were performed,and Western blotting was applied to detect the expression of ALOX5,ALOX12 and MCP-1 in the left atrial tissues.Results The diabetic group had significantly increased interventricular septal thickness but decreased LVEF than the control group(P<0.05).The collagen volume fraction of the left atrial tissue was obviously increased in the diabetes group than the control group[(7.97±1.01)%vs(3.11±0.99)%,P<0.01].The expression levels of ALOX5 and ALOX12 in the left atrium were statistically higher in the diabetes group than the control group(P<0.05),but no such difference was seen in that of of MCP-1 between the two groups(P>0.05).Conclusion The expression of ALOX5 and ALOX12 may be related to atrial inflammation and fibrosis in diabetic rabbits.
