摘 要:目的探讨巴利森苷C对氧糖剥夺/复糖复氧(OGD/R)损伤的HT22细胞氧化应激的保护作用。方法HT22细胞常规培养,随机分为对照组、模型组、依达拉奉组(100μmol/L)、低、中、高浓度组(巴利森苷C 20、40、80μmol/L)。依达拉奉和巴利森苷C干预24 h后,建立体外OGD/R模型,采用MTT法检测细胞存活率,倒置显微镜下观察细胞形态,试剂盒法测定细胞乳酸脱氢酶(LDH)、活性氧、丙二醛、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)变化,流式细胞技术膜联蛋白V-异硫氰酸荧光素/碘化丙啶双染检测细胞凋亡。结果与对照组比较,模型组细胞存活率、GSH-PX活性明显降低,LDH漏出率、活性氧、丙二醛、细胞凋亡率明显升高,差异有统计学意义(P<0.01)。与模型组比较,依达拉奉组、低浓度组、中浓度组、高浓度组LDH漏出率、活性氧明显降低,细胞存活率及GSH-PX活性明显升高(P<0.05,P<0.01),依达拉奉组、中浓度组丙二醛含量明显降低[(3.9±1.4)nmol/mg、(3.6±0.7)nmol/mg vs(5.7±1.5)nmol/mg,P<0.05],依达拉奉组、中浓度组、高浓度组细胞凋亡率明显降低,差异有统计学意义(P<0.05,P<0.01)。结论巴利森苷C对OGD/R损伤的HT22细胞氧化应激具有保护作用。Objective To investigate the protective effect of parishin C on oxidative stress in HT22 cells induced by oxygen-glucose deprivation and reperfusion(OGD/R).Methods HT22 cells were routinely cultured and randomly divided into control group,model group,edaravone group(100μmol/L),and low-,medium-and high-dose parishin C groups(20,40,80μmol/L).After the intervention of edaravone and Parishin C for 24 h,an in vitro OGD/R model was established in HT22 cells,then cell viability was detected by MTT assay,cell morphology was observed under an inverted microscope,and changes of lactate dehydrogenase(LDH),reactive oxygen species(ROS),malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)were determined with corresponding reagent kits.Cell apoptosis was detected by flow cytometry with Annexin V-FITC/PI double staining.Results Compared with the control group,the cell survival rate and GSH-PX activity were significantly decreased,and the LDH leakage rate,contents of ROS and MDA and apoptotic rate were obviously increased in the model group(P<0.01).Compared with the model group,the edaravone,and low-,medium-,and high-dose groups had notably reduced LDH leakage rate and ROS content and elevated cell survival rate and enhanced GSH-PX activity(P<0.05,P<0.01).The MDA content in the edaravone group and the medium-dose group was statistically decreased than that in the model group(3.9±1.4 nmol/mg and 3.6±0.7 nmol/mg vs 5.7±1.5 nmol/mg,P<0.05).The apoptotic rate was significantly lower in the edaravone group and the medium-and high-dose parishin C groups than the model group(P<0.05,P<0.01).Conclusion Parishin C shows a protective effect on oxidative stress in OGD/R-injured HT22 cells.
