摘  要:以黄酮类化合物的得率为考察指标,在单因素实验基础上通过正交实验优化微波辅助提取柚子内囊衣黄酮的工艺条件。实验结果表明柚子内囊衣黄酮的最佳提取工艺条件为:乙醇浓度为70%、液料比为15∶1、微波功率为240 W、微波时间为15 min。在此最佳工艺条件下,柚子内囊衣黄酮的得率为14.15 mg/g;以DPPH·清除率为考察指标,探讨最优提取工艺条件下获得的柚子内囊衣黄酮的体外抗氧化活性。结果显示在0.05~0.8 mg/mL的浓度范围内柚子内囊衣黄酮对DPPH·的清除率随其浓度增大而增强(IC50=0.11 mg/mL),表明其具有一定的抗氧化效果,但弱于VC。本研究可为其他药食兼用植物来源的黄酮类化合物的高效提取及综合利用开发提供一定的参考。Taking the extraction yield of flavonoids as an evaluation index, the conditions of microwave-assisted extraction of flavonoids from pomelo inner capsule coats were optimized by orthogonal experiment on the basis of single factor experiment. The results showed that the optimum extraction conditions of flavonoids from pomelo inner capsule coats were as follows: ethanol concentration 70 %, ratio of liquid to material 15∶1, microwave power 240 W, microwave time15min. Under the optimum conditions, the extraction yield of flavonoids from pomelo inner capsule coats was 14.15 mg/g;Using DPPH· clearance rate as an investigation index, the antioxidant activity of flavonoids in pomelo inner capsule coats obtained under the optimal extraction conditions in vitro was studied. The results showed that in the concentration range of 0.05~0.8 mg/mL, the scavenging rate of flavonoids in pomelo inner capsulecoats on DPPH· increased with the increase of its concentration(IC50=0.11 mg/mL), indicating that it had a certain antioxidant effect, but was weaker than VC. This study could provide a reference for the efficient extraction and comprehensive utilization of flavonoids from other medicinal and edible plants.
