摘　　要：目的 探究苗药吉祥草血清对气道平滑肌细胞增殖及白细胞介素(IL)-1β/环氧合酶(COX)-2信号通路的作用机制。方法 制备苗药吉祥草含药血清,分离并鉴定气道平滑肌细胞,把细胞分为空白组、低剂量组、中剂量组和高剂量组。CCK-8检测细胞的增殖能力;流式细胞仪检测细胞的凋亡情况;酶联免疫吸附试验(ELISA)检测细胞培养上清液中IL-1β和COX-2的含量;Western印迹检测细胞中IL-1β和COX-2蛋白表达。结果 鉴定发现本实验所分离培养的细胞为ASMCs。与空白组相比,低剂量组、中剂量组和高剂量组细胞在48和72 h时ASMCs细胞增殖能力、IL-1β和COX-2明显降低,凋亡率显著增加,且呈剂量依赖性(P<0.05)。结论 苗药吉祥草血清可抑制气道平滑肌细胞增殖,促进其凋亡;对细胞中炎性因子IL-1β和COX-2表达也能进行有效的抑制。Objective To investigate the mechanism of the effect of the serum of the Miao medicine auspicious grass on the proliferation of airway smooth muscle cells and the interleukin(IL)-1β/cyclooxygenase(COX)-2 signaling pathway.Methods The serum containing auspicions grass was prepared and the airway smooth muscle cells were isolated and identified. The cells were divided into blank group, low-dose, medium-dose and high-dose group. CCK-8 was used to detect cell proliferation;flow cytometry was used to detect cell apoptosis;enzyme linked immunosorbent assay(ELISA) was used to detect the content of IL-1β and COX-2 in cell culture supernatant;Western blot was used to detect the protein expression of IL-1β and COX-2.Results The cultured cells were identified as ASMCs.Compared with the blank group, the cell proliferation, expressions of IL-1β and COX-2 of ASMCs in the low-dose, middle-dose and high-dose groups were significantly reduced at 48 h and 72 h, and the apoptosis rate were significantly increased, which showed a dose-dependent(P<0.05).Conclusions The serum of Miao medicine auspicions grass could inhibit the proliferation and promote apoptosis of airway smooth muscle cells, and it also inhibit the expression of inflammatory factors IL-1β and COX-2 in the cells.
