摘 要:目的:观察防风水煎液和防风粗多糖的抗抑郁药效,探讨其作用机制。方法:采用悬尾实验和强迫游泳实验建立行为绝望小鼠抑郁模型,评价防风水煎液和防风粗多糖的抗抑郁药效。采用孤养结合慢性温和不可预知性应激(CUMS)刺激方式建立大鼠抑郁模型,观察防风粗多糖对模型大鼠体质量、糖水偏好实验、旷场实验、强迫游泳实验的影响;ELISA法检测大鼠血清促肾上腺素皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)、皮质酮(CORT)含量和海马组织色氨酸(TRP)、色氨酸羟化酶(TPH)、5-羟色胺(5-HT)、单胺氧化酶(MAO)、5-羟吲哚乙酸(5-HIAA)含量;免疫组织化学法观察大鼠海马组织糖皮质激素受体(GR)和5-羟色胺1A受体(5-HT1AR)的阳性表达。结果:防风水煎液和防风粗多糖能显著减少行为绝望小鼠在悬尾实验和强迫游泳实验中的不动时间(P<0.05或P<0.01)。与正常对照组比较,CUMS模型对照组大鼠体质量、糖水偏好率、旷场实验水平运动距离与垂直站立次数显著减少(P<0.01),强迫游泳不动时间显著增加(P<0.01),血清CRH、ACTH、CORT含量显著升高(P<0.01),海马组织TRP、TPH、5-HT含量显著降低(P<0.01),MAO、5-HIAA含量明显升高(P<0.05或P<0.01),海马CA1、CA3和DG区GR与5-HT1AR的阳性表达显著降低(P<0.01);与模型对照组比较,防风粗多糖可明显增加大鼠体质量、糖水偏好率、旷场实验水平运动距离与垂直站立次数(P<0.05或P<0.01),显著减少大鼠强迫游泳不动时间(P<0.01);明显降低大鼠血清CRH、ACTH、CORT含量(P<0.05或P<0.01);明显升高大鼠海马组织TRP、TPH、5-HT含量(P<0.05或P<0.01),明显降低海马组织MAO、5-HIAA含量(P<0.05或P<0.01);不同程度增加大鼠海马组织GR和5-HT1AR的阳性表达(P<0.05或P<0.01)。结论:防风水煎液和防风粗多糖均具有抗抑郁作用,防风粗多糖是防风抗抑郁的重要部位,其作用机制可能与增加海马组织GR的阳性表达�Objective:To observe the antidepressant effects of SAPOSHNIKOVIAE RADIX decoction and its crude polysaccharides and explore its mechanism.Methods:The mouse model of depression with behavioral despair was established by tail suspension experiment and forced swimming experiment,and the antidepressant effects of SAPOSHNIKOVIAE RADIX decoction and its crude polysaccharides were evaluated.The mouse model of depression was established by combining solitary nourishment with chronic mild and unpredictable stress stimμLation(CUMS).The effects of crude polysaccharides from SAPOSHNIKOVIAE RADIX on the body weight of the mouse model,sugar water preference experiment,open field experiment,and forced swimming experiment were observed.The content of corticotropin-releasing hormone(CRH),adrenocorticotropic hormone(ACTH),and corticosterone(CORT)in the serum and tryptophan(TRP),tryptophan hydroxylase(TPH),5-hydroxytryptamine(5-HT),monoamine oxidase(MAO),and 5-hydroxyindoleacetic acid(5-HIAA)in the hippocampus of mice were determined by enzyme-linked immunosorbent assay(ELISA).The positive expressions of the glucocorticoid receptor(GR)and 5-hydroxytryptamine 1 Areceptor(5-HT1 AR)inthe hippocampus of mice were observed by the immunohistochemical method.Results:The SAPOSHNIKOVIAE RADIX decoction and its crude polysaccharides significantly reduced the immobility time of mice with behavioral despair in the tail suspension experiment and the forced swimming experiment(P<0.01,P<0.05).As compared with the normal control group,the body weight,sugar water preference percentage,horizontal exercise distance,and vertical standing times of open field experiment in the CUMS model control group were significantly decreased(P<0.01),the immobility time of forced swimming was significantly increased(P<0.01),the content of CRH,ACTH,and CORT in the serum were significantly increased(P<0.01),the content of TRP,TPH,and 5-HT in the hippocampus were significantly decreased(P<0.01),and the content of MAO and 5-HIAA in the hippocampus were significantly in
