摘　　要：目的:采用一测多评法同时测定蒙药制剂胡日查-6丸中羟基红花黄色素A、没食子酸、鞣花酸、去氢木香内酯和木香烃内酯的含量。方法:采用Shim-pack VP-ODS C18色谱柱(250 mm×4.6 mm,5μm),以甲醇(A)-乙腈(B)-0.5%磷酸水溶液(C)为流动相,流速为1.0 mL·min^(-1)进行梯度洗脱,柱温为25℃,检测波长选择为403 nm(羟基红花黄色素A)、271 nm(没食子酸)、254 nm(鞣花酸)、225 nm(去氢木香内酯和木香烃内酯);以羟基红花黄色素A为参照物,建立羟基红花黄色素A与没食子酸、鞣花酸、去氢木香内酯和木香烃内酯的相对校正因子,测定胡日查-6丸中5个成分的含量。结果:羟基红花黄色素A、没食子酸、鞣花酸、去氢木香内酯和木香烃内酯分别在24.57~196.58、78.45~627.60、101.03~808.20、11.98~95.84、5.36~42.88μg·mL-1范围内线性关系良好;平均加样回收率(RSD)分别为98.6%(1.1%)、99.7%(0.75%)、97.8%(1.4%)、96.6%(1.1%)、102.3%(1.2%);一测多评法所得结果与外标法无显著差异。结论:建立的一测多评法可作为简便准确的质量评价模式用于蒙药制剂胡日查-6丸中的多指标质量评价。Objective:To establish a method for simultaneous determination of hydroxy safflower yellow A,gallic acid,ellagic acid,dehydrocostus lactone and costunolide in Mongolian medicine preparation Huricha-6 pills by quantitative analysis of multi-components by single marker(QAMS).Methods:The analysis was performedon a Shim-pack VP-ODS C18column(250 mm×4.6 mm,5μm),with the mobile phase comprising of methanol(A)-acetonitrile(B)-0.5%phosphoric acid(C)solution flowing at 1.0 mL·min^(-1)by gradient elution.The column temperature was set at 25℃,and the detection wavelengths were set at 403 nm(hydroxy safflower yellow A),271 nm(gallic acid),254 nm(ellagic acid)and 225 nm(dehydrocostus lactone and costunolide).With hydroxy safflower yellow A as the internal reference substance,the relative correction factors(RCFs)of gallic acid,ellagic acid,dehydrocostus lactone and costunolide to hydroxy safflower yellow A were calculated and established.The RCFs were used for the contents analysis of Huricha-6 pills.Results:The standard curves of hydroxy safflower yellow A,gallic acid,ellagic acid,dehydrocostus lactone and costunolide showed good linearity in 24.57-196.58,78.45-627.60,101.03-808.2,11.98-95.84 and 5.36-42.88μg·mL-1,respectively.The average recoveries and the corresponding RSD were 98.6%(1.1%),99.7%(0.75%),97.8%(1.4%),96.6%(1.1%)and 102.3%(1.2%),respectively.There were no significant differences between the results obtained by QAMS and those obtained by external standard method.Conclusion:The QAMS method is feasible and credible,and can be used for the multi-index quality evaluation of Mongolian medicine preparation Huricha-6 pills.
