摘  要:目的分析微量元素硒对子痫前期合并胆汁淤积患者内皮祖细胞增殖及迁移活性的影响。方法收集34例子痫前期合并胆汁淤积患者的空腹晨间静脉血60 ml,分离并培养内皮祖细胞,分别采用MTT比色法、Transwell小室检测,不同浓度的微量元素硒对内皮祖细胞增殖及迁移活性的影响,采用RT-PCR、Western blot法测定不同浓度的微量元素硒对Caspase-3 mRNA、Caspase-3蛋白表达量的影响。结果与对照组比较,随着微量元素硒浓度增加,A值逐渐上升,且16 mmol/L微量元素组﹥8 mmol/L微量元素组﹥4 mmol/L微量元素组﹥对照组,差异有统计学意义(P<0.05)。高倍镜视野下,16 mmol/L微量元素组迁移细胞数﹥8 mmol/L微量元素﹥4 mmol/L微量元素﹥对照组,差异有统计学意义(P<0.05),提示随着微量元素硒浓度的增加,内皮祖细胞迁移能力增强。对照组比较,不同浓度条件下,caspase-3 mRNA表达均显著下降,且随浓度增加,caspase-3 mRNA表达逐渐降低,16 mmol/L微量元素组Caspase-3 mRNA﹤8 mmol/L微量元素组﹤4 mmol/L微量元素组﹤对照组,差异有统计意义(P<0.05)。与对照组比较,不同剂量的硒均使caspase-3蛋白表达量显著下降,且随剂量增加,caspase-3蛋白表达量逐渐降低。16 mmol/L微量元素组Caspase-3蛋白表达量﹤8 mmol/L微量元素﹤2 mmol/L微量元素﹤对照组,差异有统计意义(P<0.05)。结论子痫前期合并胆汁淤积患者,微量元素硒或可促进内皮祖细胞增殖,提升其迁移活性,下调Caspase-3 mRNA及Caspase-3蛋白表达量。Objective To analyze the effect of trace element selenium on the proliferation and migration of endothelial progenitor cells in patients with preeclampsia and cholestasis.Methods We collected 60 ml of fasting morning venous blood from 34 patients with preeclampsia complicated with cholestasis,isolated and cultured endothelial progenitor cells.MTT colorimetry and Transwell chamber were used to detect the effects of different concentrations of trace element selenium on the proliferation and migration of endothelial progenitor cells.RT-PCR and Western blot were used to determine the effects of different concentrations of trace element selenium on the expression of Caspase-3 mRNA and Caspase-3 protein.Results Compared with the control group,with the increase of trace element selenium concentration,the A value gradually increased,and the A value of trace element selenium 16mmol/L group was higher than that of trace element selenium 8mmol/L>that of trace element selenium 4mmol/L>that of the control group,the difference was statistically significant(P<0.05).At high magnification,the number of migrated cells in the trace element selenium 16mmol/L group was higher than that in the trace element selenium 8mmol/L>that in the trace element selenium 4mmol/L>that in the control group,and the difference was statistically significant(P<0.05).Compared with the control group,caspase-3 mRNA expression decreased significantly at different concentrations,and caspase-3 mRNA expression gradually decreased with the increase of concentration.In the trace element selenium 16mmol/L group,caspase-3 mRNA<trace element selenium 8mmol/L<trace element selenium 4mmol/L<the control group,the difference was statistically significant(P<0.05).Compared with the control group,the expression of caspase-3 protein decreased significantly with different doses of selenium,and the expression of caspase-3 protein decreased gradually with the increase of the dose.The expression of Caspase-3 protein in the trace element selenium 16mmol/L group was lower than t
