摘  要:为探讨原人参二醇(PPD)、人参皂苷CK和原人参三醇(PPT)对肾病综合征(NS)中足细胞损伤的保护作用,本研究采用阿霉素(ADR)诱导足细胞损伤。通过CCK8及细胞划痕、Transwell实验分析,结果表明三种人参皂苷可以改善ADR损伤后足细胞的增殖活力并抑制其迁移能力。Western blot及细胞免疫荧光实验结果表明PPD、人参皂苷CK和PPT均可以通过降低Desmin、NF-κB P65、p-NF-κB P65、p-IκBα及升高IκBα的表达水平缓解足细胞损伤,并且可以促进GR的表达及其核转位,其中PPD的作用效果较好。计算机分子对接结果显示PPD、人参皂苷CK和PPT与GR均具有一定的结合活性,但PPD与GR的结合活性最优。因此,PPD、人参皂苷CK和PPT均可以在一定程度上改善ADR诱导的足细胞损伤,其中PPD在保护足细胞损伤方面具有一定的优势。In order to explore the protective effects of Protopanaxadiol(PPD),Ginsenosides CK and Protopanaxatriol(PPT)on podocytes injury in nephrotic syndrome(NS),the podocytes injury model is established by stimulating podocytes injury with adriamycin(ADR).The results show that the three ginsenosides can improve the proliferation and migration of ADR-induced podocytes through CCK8,cell scratch and transwell method.The results of Western blot and immunofluorescence show that PPD,Ginsenosides CK and PPT can alleviate podocytes injury by reducing the expression levels of Desmin,NF-κB P65,p-NF-κB P65 and p-IκBαproteins and increasing the expression level of IκBαproteins,and promote the GR expression and its nuclear translocations.Among them,PPD has a better effect.In addition,the results of molecular docking reveal that PPD,Ginsenosides CK and PPT are able to bind GR,and PPD exhibits the best binding activity.Therefore,PPD,Ginsenoside CK and PPT can ameliorate podocyte injury induced by ADR in some way,and PPD has more advantages in protecting podocyte injury.
