摘 要:目的 根据系统药理学方法预测天山堇菜治疗2型糖尿病的作用机制,并通过动物实验验证降糖效果。方法 通过数据库检索和文献挖掘获得天山堇菜活性成分、作用靶点及2型糖尿病相关的疾病靶点,运用Cytoscape 3.7.2软件绘制活性成分-靶点网络图,构建靶点PPI网络图,进行GO分析及KEGG通路分析,同时建立糖尿病小鼠模型验证不同浓度的天山堇菜提取物降糖效果。结果 筛选出10个活性化合物,干预2型糖尿病的关键靶点35个,多个通路与2型糖尿病发病机制相关。动物实验表明,天山堇菜提取物对糖尿病小鼠具有降糖作用,对于正常小鼠血糖没有影响。结论 本文采用系统药理学方法预测了天山堇菜的作用机制,表明天山堇菜通过多靶点、多通路产生抗2型糖尿病的作用,该作用通过动物实验得到了验证,为天山堇菜的进一步开发提供了理论基础。Objective To predict the mechanism of Viola tianshanica Maxim in the treatment of type 2 diabetes by systems pharmacology, the hypoglycemic effect was verified by animal experiments. Methods The active components, targets and disease targets related to type 2 diabetes mellitus of Viola tianshanica were obtained by database search and literature mining. Cytoscape software(version 3.7.2) were used to build a network diagram between active components and targets. “type 2 diabetes mellitus” targets were used to build a PPI network, Go analysis and KEGG pathway analysis were carried out, and diabetic mice models were established to evaluate the hypoglycemic effect of different concentrations of Viola tianshanica extract. Results Ten kinds of active compounds were screened out and 35 key targets were involved in the pathogenesis of type 2 diabetes. The extract of Viola tianshanica has hypoglycemic effect on diabetic mice, but has no effect on normal mice. Conclusion The systems pharmacology method is used to predict the mechanism of action of Viola tianshanica Maxim, it has the effect of anti-type 2 diabetes through multiple targets and pathways. and the experimental results show that the prediction method is reliable, which provide a theoretical basis for the development of Viola tianshanica Maxim.
