摘  要:目的:建立测定糖降肾康颗粒中总木脂素苷含量的方法。方法:采用双波长紫外分光光度法,在200~400 nm范围内对牛蒡苷对照品溶液、绿原酸对照品溶液以及糖降肾康颗粒供试品溶液进行扫描,在牛蒡苷对照品溶液紫外扫描图中确定最大吸收波长280 nm为测定波长λ1;在绿原酸对照品溶液紫外扫描图中,确定检测波长(λ_(1)=280 nm)的等吸收点波长350 nm为参比波长λ_(2);以牛蒡苷为对照品,测定糖降肾康颗粒中总木脂素苷的含量。结果:该方法的精密度、重现性、稳定性良好,在0.00795~0.07155 mg·mL^(-1)浓度范围内,牛蒡苷的浓度与吸光度差值呈线性相关,r=0.9999,平均回收率为100.8%,RSD为1.04%(n=6);三批糖降肾康颗粒中含总木脂素苷以牛蒡苷计,均不少于20%。结论:采用等吸收双波长紫外分光光度法测定总木脂素含量方法操作简便,准确度、精密度、稳定性良好,可作为糖降肾康颗粒中总木脂素苷的含量测定和质量控制方法。Objective To establish a determination method for the content of total lignosides in Tangjiangshenkang granules.Method By two-wavelength ultraviolet spectrophotometry(TWUS),scan arctiin control solution,chlorogenic acid control solution and tangjiangshenkang granule test solution in the range of 200-400nm.In the ultraviolet scanning diagram of arctiin reference solution,the maximum absorption wavelength of 280nm was determined as the determination wavelengthλ1.Determine the detection wavelength in the ultraviolet scanning diagram of chlorogenic acid reference solution(λ_(1)=280nm),and 350nm is the reference wavelengthλ_(2);The content of total lignosides in tangjiangshenkang granules was determined with arctiin as the reference substance.Result The precision,accuracy,and durability of this method were fine.The concentration of arctiin was linearly correlated with the absorbance difference in the range of 0.00795-0.07155 mg·mL^(-1)(r=0.9999).The average recovery of arctiin was 100.8%,and the RSD value was 1.04%(n=6).Calculated as arctiin,three batches of tangjiangshenkang granules contain no less than 20%of total lignosides.Conclusion The method has the advantages of simple operation,good accuracy,precision and reliable stability.It can be used as the content determination and quality control method of total lignosides in Tangjiangshenkang granules.
