摘  要:研究旨在探索快、慢羽鸡对禽白血病感染的天然免疫反应差异。试验采用J亚群禽白血病病毒(ALV-J)SCAU-HN06株接种快羽鸡和慢羽鸡,从接种病毒后第1天开始,每隔1天采样至第15天,采用实时荧光定量和ELISA检测快、慢羽鸡干扰素α(IFN-α)、白细胞介素12(IL-12)、肿瘤坏死因子(TNF-α)及抗病毒基因的表达情况。结果显示:慢羽鸡较快羽鸡极显著易感ALV-J(P<0.01);接种病毒后env基因的相对表达量在快羽鸡体内上升较慢羽鸡缓慢,表达峰值较慢羽鸡小;慢羽鸡接种病毒第1天IFN-α、TNF-α和IL-12蛋白表达量高于快羽鸡,随后IFN-α、TNF-α、IL-10和IL-12的蛋白表达量呈降低趋势,第11天表达量降低不显著(P>0.05);快羽鸡在接种病毒后第1天IFN-α、TNF-α和IL-12蛋白表达量升高,第13天极显著降低(P<0.01);快羽鸡抗病毒基因OAS、PKR、MX表达量显著或极显著高于慢羽组且强度强(P<0.05或P<0.01)。综上所述,慢羽鸡免疫系统比快羽鸡更早被ALV-J突破,慢羽鸡抗ALV-J的天然免疫反应比快羽鸡迟钝且强度较弱。The purpose of the study was to explore the difference of natural immune response of fast-feathered and slowfeathered chicken to avian leukemia virus infection.Subgroup J avian leukemia virus(ALV-J)SCAU-HN06 strain was used to challenge fast-feathered and slow-feathered chicken.Samples were collected every other day from the day of challenge until the15th day.Real-time fluorescence quantification and ELISA were used to detect virus infection and expression of interferon α(IFN-α),interleukin-12(IFN-12),tumor necrosis factor(TNF-α)and antiviral genes in fast-and slow-feathered chicken.The results showed that slow-feathered chicken was highly susceptible to ALV-J than fast-feathered chicken(P<0.01);After ALV-J challenge,the relative expression of env gene in fast-feathered chicken increased more slowly than that in slow-feathered chicken,and its peak expression was smaller than that in slow-feathered chicken;The protein expression levels of IFN-α,TNF-α and IL-12 in slow-feathered chicken were higher than those in fast-feathered chicken on the day of ALV-J challenge.The protein expression levels of IFN-α,TNF-α,IL10 and IL-12 of slow-feathered chicken showed a decreasing trend after ALV-J challenge,but decreased rapidly at the 11st day after ALV-J challenge(P>0.05).The protein expression levels of IFN-α,TNF-αand IL-12increased in fast feathered chicken on the day of ALV-J challenge and decreased significantly at the 13th day after ALV-J challenge(P<0.01);The frequency of significant or extremely significant differences in expression levels of OAS,PKR and MX genes were higher in fast-feathered chicken than those in slow-feathered chicken(P<0.05 to P<0.01),and the intensity was stronger.In conclusion,the immune system of slow-feathered chicken was broken through by ALV-J earlier than that of fast-feathered chicken,and the natural immune response of slow-feathered chicken against ALV-J was slower and weaker than fast-feathered chicken.
