摘 要:为探究长春市某观赏鱼水产养殖基地患病红鲫的病原及特性,采用菌落特性、理化特征、16S rDNA以及gyrB测序结果相结合的方法对病鱼肝脏组织中分离得到的1株优势菌株CI0701进行分析鉴定;同时,进行回归动物试验、药物敏感性试验、毒力基因及耐药基因检测。鉴定结果表明,该菌与豚鼠气单胞菌(Aeromonas caviae)聚类,且同源性高达99%以上;后续试验结果发现CI0701具有较强致病性,对红鲫和斑马鱼的半数致死量(LD_(50))分别为4.3×10^(3)、5.56×10 CFU/尾;该菌对红霉素、四环素、新霉素、氨苄西林、环丙沙星等12种药物耐药,属于多重耐药菌;携带溶血素(hly)、气溶素(aer)、细胞毒性肠毒素(act)、细胞兴奋性肠毒素(alt)、脂肪酶(Lip)、丝氨酸蛋白酶(Ser)和鞭毛蛋白(fla)7种毒力基因;携带氨基糖苷类[ant(3″)-Ia]、四环素类(tetA)、大环内酯类(ermB)、喹诺酮类(qnrS)和β-内酰胺类(bla_(TEM))5种耐药基因。本研究可以为红鲫携带豚鼠气单胞菌的分离鉴定以及治疗提供一定的理论基础。In order to explore the pathogen and characteristics of the diseased red crucian carp in an ornamental fish aquaculture base in Changchun City, the combined method with colony characteristics, biochemical identification, 16 S rDNA and gyrB sequencing results was used to identify a dominant strain CI0701 isolated from the liver tissue of diseased fish. Regression animal test, drug susceptibility test, virulence and resistance genes detection were also performed on the strain. The identification results showed that the bacterium was clustered with the bacterium Aeromonas caviae, with a 99% homology. The results of the follow-up test found that CI0701 was highly pathogenic, with LD_(50) on the red crucian carp and zebrafish being 4.3×10^(3) and 5.56×10 CFU/tail, respectively. The bacterium was resistant to 12 kinds of drugs, such as erythromycin, tetracycline, neomycin, ampicillin, ciprofloxacin, and so on. Because it carried 7 virulence genes, such as hly, aer, act, alt, Lip, Ser, and fla;and 5 drug resistance genes, such as ant(3″)-Ia, tetA, ermB, qnrS, and bla_(TEM). This study can provided theoretical support for the isolation, identification and treatment of Aeromonas caviae carried by crucian carp.
