摘 要:目的探讨柯孜木克颗粒(KZMK)对代谢综合征(MetS)模型大鼠炎性状态的影响。方法将36只SD大鼠随机分为正常组(等体积生理盐水),模型组(等体积生理盐水),KZMK高、中、低剂量组(70.38,35.19,17.6mg/kg)及二甲双胍组(3mg/kg),各6只。正常组大鼠饲以普通饲料,其余组饲以高盐高脂高糖饲料12周,以复制MetS大鼠模型。喂饲8周后,6组大鼠灌胃给予相应药物或生理盐水,每日1次,连续4周。采用苏木素-伊红染色,观察大鼠肝脏组织病理形态;采用酶联免疫吸附(ELISA)法检测大鼠血清肿瘤坏死因子-α(TNF-α)水平;采用免疫组织化学法检测大鼠肝脏组织核因子κB(NF-κB)表达水平。结果模型组大鼠肝小叶结构不清晰,肝细胞松散、肿胀、紊乱,炎性细胞浸润;KZMK各剂量组上述病理形态均有所改善。与模型组比较,KZMK各剂量组大鼠血清TNF-α水平均显著降低(P<0.05);KZMK各剂量组肝脏组织NF-κB表达水平均显著降低(P<0.05)。结论KZMK对MetS模型大鼠肝脏组织病理形态有一定的改善作用,作用机制可能与抑制NF-κB的活化,减轻炎性状态有关。Objective To investigate the effect of Kezimuke Granules(KZMK)on inflammatory state in rats with metabolic syndrome(MetS).Methods Thirty-six SD rats were randomly divided into normal group(equal volume of normal saline),model group(equal volume of normal saline),KZMK high-,medium-and low-dose groups(70.38,35.19,17.6mg/kg)and metformin group(3mg/kg),with six rats in each group.The rats in the normal group were fed with normal diet,and the rats in the other groups were fed with high-salt,high-fat and high-sugar diet for 12 weeks to replicate the MetS rat model.After eight weeks of feeding,rats in six groups were given corresponding drugs or normal saline by gavage once a day for four weeks.The histopathological morphology of liver were observed by hematoxylin-eosin(HE)staining.The serum level of tumor necrosis factor-α(TNF-α)was detected by the enzyme-linked immunosorbent assay(ELISA).The expression level of nuclear factor-κB(NF-κB)in rat liver tissue was detected by the immunohistochemistry.ResultsIn the model group,the structure of the hepatic lobule was not clear,the hepatocytes were loose,swollen and disorganized with inflammatory cell infltration.However,the above pathological morphology was improved in KZMK high-,medium-and low-dose groups.Compared with those in the model group,the serum TNF-αlevel decreased significantly in KZMK high-,medium-and low-dose groups(P<0.05),the expression level of NF-κB in liver tissue decreased significantly in KZMK high-,medium-and low-dose groups(P<0.05).Conclusion KZMK show some ameliorative effect on the histopathological changes of the liver in MetS model rats,and its mechanism may be related to inhibiting the activation of NF-κB and reducing the inflammatory state.
