摘  要:目的探讨不同剂量的柑橘类多甲氧基黄酮对小鼠的急性毒性。方法分别给予氯化钠溶液和多甲氧基黄酮灌胃给药后,观察小鼠的活动、饮食、饮水、体质量、死亡等情况,用加权回归法(weighted regression method,Bliss)计算半数致死量(median lethal dose,LD_(50))及95%置信区间。结果多甲氧基黄酮对小鼠的毒性实验结果表明多甲氧基黄酮对存活小鼠饮食、饮水及体质量无明显影响。通过Bliss法计算得多甲氧基黄酮的LD_(50)为3.561 g/kg,表现出一定的毒性,LD_(50)的95%置信区间为2.258~6.451 g/kg;回归方程为Y(Probit)=-1.034+1.874 lgD。结论多甲氧基黄酮具有一定的毒性,且脏器指数与病理观察显示毒性主要表现在血液血管方面,主要作用在肝、脾和肾脏,其毒性与给药剂量相关,在一定剂量范围内可安全使用。Objective The acute toxicity of citrus polymethoxy flavones in mice was studied.Methods Activities,diet,drinking water,body weight,death and other conditions of mice were observed after gavage of normal saline and polymethoxy flavones,respectively.Median lethal dose(LD_(50))was calculated by Weighted regression method(Bliss)and 95% confidence interval.Results The toxicity test of polymethoxy flavones on mice showed that polymethoxy flavones had no obvious effect on diet,drinking water or body weight of mice.The value of LD_(50)of polymethoxyl flavones calculated by Bliss method was 3.561 g/kg,showing certain toxicity,and the 95% confidence interval of LD_(50)was 2.258-6.451 g/kg.The regression equation was Y(Probit)=-1.034+1.874 lgD.Conclusions Polymethoxy flavones have certain toxicity,and organ index and pathological observation show that the toxicity manifested in blood vessels,mainly acting on liver,spleen and kidney.Its toxicity is related to dose,and it can be used safely within a certain dose range.
