摘  要:目的基于NF-κB信号通路探讨牛膝总皂苷治疗膝骨性关节炎滑膜炎症的机制。方法选择SPF级SD大鼠60只,随机分成对比组、模型组、牛膝总皂苷低剂量组、牛膝总皂苷中剂量组、牛膝总皂苷高剂量组,每组12只。除对比组外,其他各组大鼠在关节两侧进针,从第1天开始每间隔2 d分别注射0.3 mL 5%木瓜蛋白酶水溶液到大鼠膝关节腔内,比较5组大鼠关节炎症指标。结果与对比组比较,模型组的膝关节肿胀程度较严重,miR-19α的基因表达水平较低,而SOX9、NF-κB的基因水平表达较高,使用牛膝总皂苷的3组,根据剂量的增加肿胀水平依次降低,miR-19α基因表达水平依次增高,而SOX9以及NF-κB的基因表达水平和蛋白表达水平依次降低。结论牛膝总皂苷能减轻骨关节炎的炎症症状,并且能通过上调miR-19α的表达抑制SOX9、NF-κB的表达水平,保护软骨细胞的凋亡。Objective Based on the NF-κB signaling pathway to explore the therapeutic mechanism of total saponins of achyranthes bidentata in the treatment of synovial inflammation of knee osteoarthritis.Methods Sixty SPF SD rats were selected and randomly divided into control group,model group,low-dose achyranthes bidentata saponin group,achyranthes bidentata mid-dose group,achyranthes bidentata saponin high-dose group,with 12 rats in each group.Except for the control group,the rats in the other groups were injected with needles on both sides of the joints.From the first day,0.3mL of 5%papain aqueous solution was injected every two days into the knee joint cavity of the rats.The rats’joint inflammation indexes of five groups was compared.Results Compared with the control group,the knee joint swelling of the model group was more serious,the gene expression level of miR-19 was lower,and the gene expression level of SOX9 and NF-κB was higher.The three groups using total saponin of achyranthes bidentata,according to the dose the level of swelling decreased successively,the expression level of miR-19 gene increased successively,while the gene expression level and protein expression level of SOX9 and NF-κB successively decreased.Conclusion Achyranthes bidentata saponins can alleviate the inflammatory symptoms of osteoarthritis,and can pass up-regulating the expression of miR-19 inhibits the expression levels of SOX9 and NF-κB,and protects chondrocytes from apoptosis.
