摘 要:为探究农林生产常用杀虫剂对周氏啮小蜂的胁迫效应,通过药膜法评估阿维菌素对周氏啮小蜂的安全性,并利用转录组测序分析阿维菌素胁迫下周氏啮小蜂的基因表达变化.结果表明:(1)田间使用浓度的阿维菌素对周氏啮小蜂的毒力作用较低,属于低风险性农药,高浓度阿维菌素胁迫对周氏啮小蜂有短期触杀作用;(2)与对照组相比,LC_(10)和LC_(30)浓度阿维菌素胁迫下,对周氏啮小蜂进行转录组测序共获得805个差异表达基因,其中33个基因表达上调,772个基因表达下调;(3)GO功能分析结果表明,差异表达基因在细胞过程、代谢过程、膜和膜成分、分子结合和催化活性方面均显著富集;(4)KEGG代谢通路分析结果显示,神经活性配体-受体相互作用以及药物代谢细胞色素P450代谢通路显著富集;(5)对细胞色素P450相关基因的RT-qPCR检测发现,与对照组相比,阿维菌素胁迫下CcCYP9AG3和CcCYP9AG4的表达水平显著上升,说明细胞色素P450基因家族尤其是CYP9家族参与了周氏啮小蜂对阿维菌素的胁迫响应.To explore the stress effect of pesticides widely applied in agriculture and forestry on Chouioia cunea Yang,the safety of avermectin,which is a kind of antibiotic insecticide,on C.cunea was assessed by the method of film bioassay.Transcriptome sequencing was used to analyze the alternation of certain gene expression of C.cunea under avermectin stress.The results showed that:(1)Because of the low toxicity of avermectin with the concentration used in the field to C.cunea,avermectin is considered to be a low risk pesticide,while high concentration stress had short-term killing effect on C.cunea;(2)Compared with the control group,a total of 805 differentially expressed genes were obtained by transcriptome sequencing of C.cunea under avermectin stress with LC_(10)and LC_(30)concentrations,of which 33 genes were up-regulated and 772 genes were down-regulated;(3)The functional analysis of Gene Ontology(GO)annotation revealed that the corresponding differentially expressed genes were enriched in cell process,metabolic process,membrane and membrane components,molecular binding and catalytic activities;(4)The analysis of KEGG metabolic pathway demonstrated that the interaction of neuroactive ligand-receptors and the cytochrome P450-induced drug metabolizing pathway were significantly enriched;(5)Detection of cytochrome P450-related genes by RT-qPCR showed that the expression levels of CcCYP9AG3 and CcCYP9AG4 were significantly higher in the avermectin stress group than in the control group,which indicated that the cytochrome P450 gene family,in particular,the CYP9 family is involved in the response of C.cunea to avermectin stress.
