摘 要:目的:研究黄芪发酵产物(HF)拮抗脂多糖诱导的小鼠急性肺损伤(ALI)的干预效果,初步探讨其作用机制,并为其开发利用提供参考依据。方法:60只BALB/c小鼠按体质量随机分成空白对照组、模型组、舒普深组(0.3 g/kg)及HF低、中、高剂量组(12.5、25、50 g/kg),每组10只,根据文献方法采用吸入脂多糖法复制小鼠ALI模型,各组处理4周后,采用近红外荧光系统对小鼠活体成像进行观察,ELISA法检测小鼠血清TNF-α、IL-6、IL-1水平,Western blot法检测肺组织TLR4的表达,并进行肺部病理学观察。结果:模型组近红外荧光活体成像荧光信号最强,各给药组呈现不同程度的减弱的荧光信号。与模型组比较,舒普深组、HF中剂量组、HF高剂量组小鼠体质量不同程度的增加(P<0.05,P<0.01);HF中剂量组的TNF-α、IL-6的水平降低(P<0.05);HF高剂量组的TNF-α、IL-1、IL-6的水平降低(P<0.01,P<0.05);HF高剂量组TLR4表达水平降低(P<0.05)。病理学研究表明,HF能够降低脂多糖诱导的ALI小鼠的肺组织损伤程度。结论:黄芪发酵产物对脂多糖诱导的小鼠急性肺损伤具有保护作用,机制可能与其抑制TLR4通路发挥抗炎作用有关。Objective:To study the effects of Astragalus fermentative product(HF)on lipopolysaccharide-induced acute lung injury(ALI)in mice,and to explore its mechanism,thus to provide reference for its utilization.Methods:60 BALB/c mice were randomly divided into the blank control group,the model group,the Sulperazone group(0.3 g/kg)and the HF groups of low-dose,medium-dose and high-dose(12.5 g/kg,25 g/kg,50 g/kg)according to body weight,with 10 mice in each group.ALI model was replicated by inhalation of lipopolysaccharide in mice.After 4 weeks of interventions,the in vivo imaging was observed by near-infrared fluorescence system,the serum levels of TNF-α,IL-6 and IL-1 were detected by ELISA method,and Western blot method was used to detect the expression of TLR4 in lung tissues.Results:The fluorescence signal of near-infrared fluorescence in vivo imaging was the strongest in the model group,and the other medication groups showed different degrees of fading signals.Compared with those in the model group,the body mass was increased in the Sulperazone group,the HF groups of medium-dose and high-dose(P<0.05,P<0.01);the levels of TNF-αand IL-6 were significantly lowered in the HF group of medium-dose(P<0.05);the levels of TNF-α,IL-1 and IL-6 were significantly decreased in the HF group of high-dose(P<0.05,P<0.01);the expression of TLR4 was significantly lower in the HF group of high-dose(P<0.05).Pathological research showed that HF could reduce the lung tissues damage in ALI mice induced by lipopolysaccharide.Conclusion:HF has a good protective effect on ALI induced by lipopolysaccharide,which mechanism may be related to inhibiting TLR4 pathway to play the role of anti-inflammation.
