摘  要:目的优化吸水链霉菌产雷帕霉素发酵培养基,提高雷帕霉素产量。方法利用Plackett-Burman试验设计筛选出培养基中影响雷帕霉素发酵产量的显著因素,爬坡试验确定主要因素的最适范围,响应面法确定各显著因素的最优水平。结果获得最优发酵培养基配方为:葡萄糖37.60 g/L、甘露醇30 g/L、黄豆粉28.37 g/L、硫酸铵1.25 g/L、磷酸氢二钾5 g/L、磷酸二氢钾5 g/L、L-赖氨酸1.5 g/L、复合氨基酸1.83 g/L。结论在最优发酵培养基培养下,雷帕霉素发酵水平由初始182.23 mg/L提高到279.56 mg/L,提高了53.41%。Objective To optimize rapamycin-producing Streptomyces hygroscopicus medium and improve the yield of rapamycin.Methods Plackett Burman experimental design was applied to screen out the significant factors affecting the fermentation yield of rapamycin in fermentation medium.The main factors were tested through the steepest ascent method to get the optimal range.Finally,the Response Surface Methodology was used to determine the optimal level of each significant factor.Results The optimal formulation of fermentation medium was:glucose 37.60 g/L,mannitol 30 g/L,soybean powder 28.37 g/L,ammonium sulfate 1.25 g/L,dipotassium hydrogen phosphate 5 g/L,potassium dihydrogen phosphate 5 g/L,L-lysine 1.5 g/L and compound amino acid 1.83 g/L.Conclusion Under the optimal fermentation medium,the fermentation level of rapamycin can be increased from 182.23 mg/L to 279.56 mg/L,with an increase of 53.41%.
