摘 要:目的建立高效液相色法(HPLC)一测多评法同时测定银翘感冒袋泡剂中绿原酸、甘草苷、连翘酯苷A、连翘苷、牛蒡苷的含量。方法运用HPLC,色谱柱:Shimazu Inertsil ODS-3(150 mm×4.6 mm,5μm);流动相:乙腈-0.2%磷酸,梯度洗脱;体积流量:1.0 ml/min;柱温35℃;检测波长为220 nm。通过3种校正方法分别建立各成分与连翘酯苷A的相对校正因子,计算含量,同时采用外标法和回归方程法测定含量,并对3种校正因子所得结果的差异进行比较,从而对一测多评法的准确性和可行性加以验证。结果绿原酸、甘草苷、连翘酯苷A、连翘苷、牛蒡苷分别在83.77~670.20,6.36~50.90,41.45~331.60,9.15~73.20,31.50~252.00μg/ml范围内线性关系良好(r≥0.9998),平均加样回收率(RSD)分别为95.4%(0.32%),100.1%(0.96%),98.0%(0.18%),100.7%(1.37%),99.0%(0.57%)。基于3种校正法的一测多评法所得结果与外标法、回归方程法接近。结论该方法简便准确,可用于银翘感冒袋泡剂的质量控制。Objective To establish an HPLC-QAMS method for the simultaneous determination of chlorogenic acid,liquiritin,forsythia glycoside A,forsythia glycoside and burdock glycoside in Yinqiao Ganmao Tea Bags.Methods The separation was carried out on a Shimazu Inertsil ODS-3 column(150 mm×4.6 mm,5μm)using acetonitrile-0.2%phosphoric acid as the mobile phase for gradient elution.The flow rate was 1.0 ml/min,the detection wavelength was set at 220 nm and the column temperature was 35℃.Three correction methods were used to establish the relative correction factors between each constituent and forsythoside A,respectively,then to calculate the amount of each constituent and finally to achieve QAMS.At the same time,the external standard method and the regression equation method were used to determine the amounts of the above 5 constituents.The difference between the calculated values and real data of the three factors was compared and the accuracy and feasibility of QAMS were validated.Results Chlorogenic acid,liquiritin,forsythia-glycoside A,-forsythia gly-coside and b-urdock glyco-side showed good linear relationships within the ranges of 83.77670.20,6.3650.90,41.45331.60,9.1573.20,31.50252.00μg/ml(r≥0.9998),respectively.The average recoveries(RSDs)were 95.4%(0.32%),100.1%(0.96%),98.0%(0.18%),100.7%(1.37%)and 99.0%(0.57%),respectively.The results obtained by the QAMS based on three correction methods were closed to the external standard method and the regression equation method.Conclusion This simple and accurate method can be used for the quality control of Yinqiao Ganmao Tea Bags.
